A pervasive developmental disorder not otherwise specified (Including atypical autism) (PDD-NOS) is one of the four autistic disorders (AD) in the DSM-5 and also was one of the five disorders classified as a pervasive developmental disorder (PDD) in the DSM-IV According to the DSM-4, PDD-NOS is a diagnosis that is used for "severe or pervasive impairment in the development of reciprocal social interaction and/or verbal and nonverbal communication skills, or when stereotyped behavior, interests, and/or activities are present, but the criteria are not met for a specific PDD" or for several other disorders. PDD-NOS includes atypical autism, because the criteria for autistic disorder are not met, for instance because of late age of onset, atypical symptomatology, or subthreshold symptomatology, or all of these. Even though PDD-NOS is considered milder than typical autism, this is not always true. While some characteristics may be milder, others may be more severe.

The process by which dysfunctional mitochondria are selectively targeted for lysosome-mediated degradation otherwise known as mitophagy, requires the serine/threonine kinase PINK1 and the E3 ubiquitin ligase Parkin to occur. In the last decade the PINK1/Parkin pathway received great attention due to its importance in many physiological and pathological processes. Understanding the mechanisms by which mitochondria are selectively recognized and targeted for degradation is thus fundamental to understand and to develop therapies for many devastating diseases. Here the mechanisms at the basis of the PINK1/Parkin-mediated degradation of dysfunctional mitochondria are described.

Sensitive Clay Slope
Sensitive clays are known for producing retrogressive landslides, also called spread or flowslides. The key characteristics associated with the occurrence of these landslides on a sensitive clay slope must be assessed, and the potential retrogressive distance must be evaluated. Common risk analysis methods include empirical methods for estimating the distance of potential retrogression, analytical limit equilibrium methods, numerical modelling methods using the strength reduction technique, and the integration of a progressive failure mechanism into numerical methods. Methods developed for zoning purposes in Norway and Quebec provide conservative results in most cases, even if they don’t cover the worst cases scenario. A flowslide can be partially analysed using analytical limit equilibrium methods and numerical methods having strength reduction factor tools. Numerical modelling of progressive failure mechanisms using numerical methods can define the critical parameters of spread-type landslides, such as critical unloading and the retrogression distance of the failure. Continuous improvements to the large-deformation numerical modeling approach allow its application to all types of sensitive clay landslides.

The acquisition of iron is essential and becomes a crucial pathogenic event for opportunistic fungi. Iron is not readily available in the natural environment as it exists in its insoluble ferric form, i.e., in oxides and hydroxides. During infection, the host iron is bound to proteins such as transferrin, ferritin, and haemoglobin. As such, access to iron is one of the major hurdles that fungal pathogens must overcome in an immunocompromised host. Poly (ADP-ribose) polymerases (PARP) 1-3 are well-known multi-domain enzymes, catalysing the covalent modification of proteins, DNA, and themselves. They attach mono- or poly-ADP-ribose to targets using NAD+ as a substrate. Poly-ADP-ribosylation (PARylation) is central to the important functions of PARP enzymes in the DNA damage response and nucleosome remodelling. Activation of PARP happens through DNA binding via zinc fingers and/or the WGR domain. Powdery Mildew Resistance Loci in Vines
Grapevine (Vitis vinifera) is one of the main fruit crops worldwide, with near of 7.3 million hectares planted in 2020, but along with its economic relevance, it has been associated with diverse pathogens that affect grapevine yield, fruit, and wine quality, of which powdery mildew is the most important disease prior to harvest. Its causal agent is the biotrophic fungus Erysiphe necator, which generates a decrease in cluster weight, delays fruit ripening, and reduces photosynthetic and transpiration rates. In addition, powdery mildew induces metabolic reprogramming in its host, affecting primary metabolism. Most commercial grapevine cultivars are highly susceptible to powdery mildew; consequently, large quantities of fungicide are applied during the productive season. These pesticide applications have been associated with high exposure to it, and pesticides are associated with health problems, negative environmental impacts, and high costs for farmers. In parallel, consumers are demanding more sustainable practices during food production. Therefore, new grapevine cultivars with genetic resistance to powdery mildew are needed for sustainable viticulture, while maintaining yield, fruit, and wine quality. Two main gene families confer resistance to powdery mildew in the Vitaceae, Run (Resistance to Uncinula necator) and Ren (Resistance to Erysiphe necator), and the resistance they confer is associated with the presence of each locus since there are still no genes that alone can produce a powerful genetic resistance. Because the resistance mediated by the plant immune response is highly complex and considers the evolution and adaptation of the pathogen in parallel to that of the plant.

Plant growth and productivity are orchestrated by a network of signaling cascades involved in balancing responses to perceived environmental changes with resource availability. Vascular plants are divided into the shoot, an aboveground organ where sugar is synthesized, and the underground located root. Continuous growth requires the generation of energy in the form of carbohydrates in the leaves upon photosynthesis and uptake of nutrients and water through root hairs. Root hair outgrowth depends on the overall condition of the plant and its energy level must be high enough to maintain root growth. TARGET OF RAPAMYCIN (TOR)-mediated signaling cascades serve as a hub to evaluate which resources are needed to respond to external stimuli and which are available to maintain proper plant growth. Root hair formation further requires appropriate distribution of the phytohormone auxin, which primes root hair cell fate and triggers root hair elongation. While TOR activity partially depends on auxin signaling, auxin distribution to the root tip and the redistribution towards the root hair zone are, on the other hand, dependent on TOR activity. Together, both signaling cascades are interwoven to orchestrate proper root hair outgrowth to ensure efficient plant nutrition.

They exhibit major advantages, including stimuli-responsive properties for the “on demand” drug release and the ability for controlled release, by manipulating their internal conformation properties and their administration by different routes. Reprogramming of adult somatic cells into induced pluripotent stem cells (iPSCs) has revolutionized the complex scientific field of disease modeling and personalized therapy. Cardiac differentiation of human iPSCs into cardiomyocytes (hiPSC-CMs) has been used in a wide range of healthy and disease models by deriving CMs from different somatic cells. Unfortunately, hiPSC-CMs have to be improved because existing protocols are not completely able to obtain mature CMs recapitulating physiological properties of human adult cardiac cells. Therefore, improvements and advances able to standardize differentiation conditions are needed. Lately, evidences of an epigenetic memory retained by the somatic cells used for deriving hiPSC-CMs has led to evaluation of different somatic sources in order to obtain more mature hiPSC-derived CMs.
