Biomarker-Driven Drug Development
Biomarker-driven drug development in age of personalized medicines. A biomarker life cycle is broken down into 3 stages - discovery, translation, and qualification. Researchers review current development strategies and technologies applied at each of these stages, with emphasis on the use of real-world data as an important source of supporting evidence.

Pathogenesis of Cutaneous Melanoma
Melanoma develops from malignant transformations of the pigment-producing melanocytes. If located in the basal layer of the skin epidermis, melanoma is referred to as cutaneous, which is more frequent. However, as melanocytes are be found in the eyes, ears, gastrointestinal tract, genitalia, urinary system, and meninges, cases of mucosal melanoma or other types (e.g., ocular) may occur. The incidence and morbidity of cutaneous melanoma (cM) are constantly increasing worldwide.

Antifungal Susceptibility Testing for Fungi
Invasive fungal infections (IFIs) are associated with high mortality rates and timely appropriate antifungal therapy is essential for good outcomes. Emerging antifungal resistance among Candida and Aspergillus spp., the major causes of IFI, is concerning and has led to the increasing incorporation of in vitro antifungal susceptibility testing (AST) to guide clinical decisions.

Cardiac Tissue Engineering Systems
Cardiac tissue engineering is very much in a current focus of regenerative medicine research as it represents a promising strategy for cardiac disease modelling, cardiotoxicity testing and cardiovascular repair. Advances in this field over the last two decades have enabled the generation of human engineered cardiac tissue constructs with progressively increased functional capabilities. Numerous studies have demonstrated that the therapeutic benefits exerted by cells are mainly attributable to the release of complementary paracrine factors and the efficacy is limited as only a small percentage of transplanted cells engrafted in the infarcted tissue. Studies on animal models showed that combining cell therapy with tissue engineering techniques for the creation of cell sheets and patches, can increase stem cell survival and boost therapeutic action. Therefore, tissue engineering has been considered as a potential approach for cardiac regeneration after MI.

Fibroblast Growth Factor 23 and Osteoporosis
Osteoporosis is a chronic debilitating disease caused by imbalanced bone remodeling processes that impair the structural integrity of bone. Over the last ten years, the association between fibroblast growth factor 23 (FGF23) and osteoporosis has been studied in both pre-clinical and clinical investigations. FGF23 is a bone-derived endocrine factor that regulates mineral homeostasis via the fibroblast growth factor receptors (FGFRs)/αKlotho complex. These receptors are expressed in kidney and the parathyroid gland. Preclinical studies have supported the link between the local actions of FGF23 on the bone remodeling processes. In addition, clinical evidence regarding the effects of FGF23 on bone mass and fragility fractures suggest potential diagnostic and prognostic applications of FGF23 in clinical contexts, particularly in elderly and patients with chronic kidney disease. However, inconsistent findings exist and there are areas of uncertainty requiring exploration.

Classification of Dystonia
Dystonia is a movement disorder characterized by sustained or intermittent muscle contractions causing abnormal and often repetitive, movements, postures, or both. Dystonic movements are typically patterned, twisting, and may be tremulous. Dystonia is often initiated or worsened by voluntary action and associated with overflow muscle activation. Because of its wide clinical spectrum, dystonia is often underdiagnosed or misdiagnosed. In clinical practice, dystonia could often present in association with other movement disorders. An accurate physical examination is essential to describe the correct phenomenology.

Callous and Unemotional (CU) Traits
Behavioral disorders (BD), including disruptive, aggressive, and/or antisocial behavior, are some of the most common disorders in children and adolescents. The notion of BD used in our review is an umbrella term, including children and adolescents with a range of emotional and behavioral disorders. It includes a population with heterogeneous behavioral diagnoses such as oppositional defiant disorder (ODD), conduct disorder (CD), internalized disorders (ID), conditions often associated with aggressive and/or antisocial behavior. Retrospective research in the field notes heterogeneous developmental trajectories that could be explained by certain risk factors, including the severity of callous-unemotional (CU) traits, a constellation of emotional and personality traits in children considered as a precursor to adult psychopathy.

Phenothiazines Modified with Pyridine Ring
Azaphenothiazines are the largest and most perspective group of modified phenothiazines, and they exhibit variety of biological activities. The review sums up the current knowledge on the anticancer activity of isomeric pyridobenzothiazines and dipyridothiazines, which are modified azaphenothiazines with one and two pyridine rings, respectively, against 10 types of cancer cell lines. Some 10-substituted dipyridothiazines and even 10-unsubstituted parent compounds, such as 10H-1,9-diazaphenothiazine and 10H-3,6-diazaphenothiazine, exhibited very potent action with the IC50 values less than 1 µg/mL and 1 µM against selected cancer cell lines. The strength of the anticancer action depends both on the tricyclic ring scaffolds and the substituents at the thiazine nitrogen atom.

Therapeutic Attributes of Endocannabinoid System
In humans, various sites like cannabinoid receptors (CBR) having a binding affinity with cannabinoids are distributed on the surface of different cell types, where endocannabinoids (ECs) and derivatives of fatty acid can bind. The binding of these substance(s) triggers the activation of specific receptors required for various physiological functions, including pain sensation, memory, and appetite.

Brainstem Midline Stroke
Most medullary and pontine strokes are sharply marginated and paramedian, with the long axis being oriented in the sagittal plane. This pattern is related to the distribution of the paramedian-penetrating branches arising from the BA and distal VAs, which perforate the paramedian brainstem and never cross the midline. Lateral infarcts, in the distribution of the short circumferential arteries, are seen less frequently than paramedian lesions are. At the midbrain level, midline infarctions can be visualized, as the many involved branches are not limited to a paramedian distribution. Characteristically, brainstem vascular syndromes are also called alternating syndromes due to the presence of crossed neurological signs (ipsilateral cranial nerve signs and contralateral signs of the ascending and descending tracts) which are hallmarks of the brainstem location.
