LPA 3 Promotes Mitochondrial Homeostasis against Oxidative Stress
Lysophosphatidic acid (LPA) is a growth factor-like lipid mediator that regulates various physiological functions via activation of multiple LPA G protein-coupled receptors. Mitochondria have been suggested to be the primary origin of oxidative stress via the overproduction of reactive oxygen species (ROS). Mitochondria are responsible for producing ATP through oxidative phosphorylation (OXPHOS) and have a calcium buffering capacity for the cell. Defects in mitochondria will lead to declined antioxidant capacity and cell apoptosis. siRNA-mediated depletion of LPA3 leads to the depolarization of mitochondrial potential (ΔΨm) and cellular ROS accumulation. In addition, the depletion of LPA3 enhances cisplatin-induced cytochrome C releasing. This indicates that LPA3 is essential to suppress the mitochondrial apoptosis pathway. LPA3 is also shown to improve mitochondrial ADP-ATP exchange by enhancing the protein level of ANT2. On the other hand, LPA3 regulates calcium uptake from the ER to mitochondria via the IP3R1-VDAC1 channel. Moreover, activation of LPA3 by selective agonist OMPT rescues mitochondrial homeostasis of H2O2-induced oxidative stress cells and HGPS patient fibroblasts by improving mitochondrial ΔΨm and OXPHOS.

Pandemrix
Pandemrix is an influenza vaccine for influenza pandemics, such as the 2009 flu pandemic. The vaccine was developed by GlaxoSmithKline (GSK) and patented in September 2006. The vaccine was one of the H1N1 vaccines approved for use by the European Commission in September 2009, upon the recommendations of the European Medicines Agency (EMEA). The vaccine is only approved for use when an H1N1 influenza pandemic has been officially declared by the World Health Organization (WHO) or European Union (EU). The vaccine was initially developed as a pandemic mock-up vaccine using an H5N1 strain. Pandemrix was found to be associated with an increased risk of narcolepsy following investigations by Swedish and Finnish health authorities and had higher rates of adverse events than other vaccines for H1N1. This resulted in several legal cases. Stanford University studies suggested that narcolepsy is an autoimmune disease and that it appears to be triggered by upper airway respiratory infections.

Role of Glycosylation in Systemic Lupus Erythematosus
Glycosylation is a post-translational modification that affects the stability, structure, antigenicity and charge of proteins. In the immune system, glycosylation is involved in the regulation of ligand–receptor interactions, such as in B-cell and T-cell activating receptors. Alterations in glycosylation have been described in several autoimmune diseases, such as systemic lupus erythematosus (SLE), in which alterations have been found mainly in the glycosylation of B lymphocytes, T lymphocytes and immunoglobulins. In immunoglobulin G of lupus patients, a decrease in galactosylation, sialylation, and nucleotide fucose, as well as an increase in the N-acetylglucosamine bisector, are observed. These changes in glycoisolation affect the interactions of immunoglobulins with Fc receptors and are associated with pericarditis, proteinuria, nephritis, and the presence of antinuclear antibodies. In T cells, alterations have been described in the glycosylation of receptors involved in activation, such as the T cell receptor; these changes affect the affinity with their ligands and modulate the binding to endogenous lectins such as galectins. In T cells from lupus patients, a decrease in galectin 1 binding is observed, which could favor activation and reduce apoptosis. Furthermore, these alterations in glycosylation correlate with disease activity and clinical manifestations, and thus have potential use as biomarkers.

The vulvar area is a common site of contact dermatitis due to the thin skin, easily traversable by irritant and allergic substances. The nonkeratinized vulvar vestibule is likely to be more permeable than the keratinized portions of the vulva and thus more susceptible to exogenous topical agents. The vulva is an area of occlusion due to both its intrinsic anatomical structure and the frequent use of occlusive napkins or underwear, which increase penetration or absorption of both irritants and allergens. Furthermore, women at different ages, due to urine and feces as children and to vaginal mucosal atrophy and the increase in the vulvar pH in menopause, may have an altered barrier function and, in incontinent elderly subjects, the use of diapers may contribute to increased susceptibility to irritants and allergens.

Hunting Deeply Hidden Software Vulnerabilities
Fuzz testing is a simple automated software testing approach that discovers software vulnerabilities at a high level of performance by using randomly generated seeds. However, it is restrained by coverage and thus, there are chances of finding bugs entrenched in the deep execution paths of the program. To eliminate these limitations in mutational fuzzers, patching-based fuzzers and hybrid fuzzers have been proposed as groundbreaking advancements which combine two software testing approaches. Despite those methods having demonstrated high performance across different benchmarks such as DARPA CGC programs, they still present deficiencies in their ability to analyze deeper code branches and in bypassing the roadblocks checks (magic bytes, checksums) in real-world programs. In this research, we design DeepDiver, a novel transformational hybrid fuzzing tool that explores deeply hidden software vulnerabilities. Our approach tackles limitations exhibited by existing hybrid fuzzing frameworks, by negating roadblock checks (RC) in the program. By negating the RCs, the hybrid fuzzer can explore new execution paths to trigger bugs that are hidden in the abysmal depths of the binary. We combine AFL++ and concolic execution engine and leveraged the trace analyzer approach to construct the tree for each input to detect RCs. To demonstrate the efficiency of DeepDiver, we tested it with the LAVA-M dataset and eight large real-world programs. Overall, DeepDiver outperformed existing software testing tools, including the patching-based fuzzer and state-of-the-art hybrid fuzzing techniques. On average, DeepDiver discovered vulnerabilities 32.2% and 41.6% faster than QSYM and AFLFast respectively, and it accomplished in-depth code coverage.

Mutant p53
The p53 tumor suppressor plays a pivotal role in cancer and infectious disease. Many oncology treatments are now calling on immunotherapy approaches, and scores of studies have investigated the role of p53 antibodies in cancer diagnosis and therapy. This review summarizes the current knowledge from the preliminary evidence that suggests a potential role of p53 as an antigen in the adaptive immune response and a monitoring key role of the innate immune system, thereby speculating on the idea that mutant p53 antigens serve as a druggable target in immunotherapy. Except in a few cases, the vast majority of published work on p53 antibodies in cancer patients use wild-type p53 as the antigen to detect these antibodies and it is unclear that they can recognize p53 mutants carried by cancer patients at all. We envision that an antibody targeting a specific mutant p53 will be effective therapeutically against the cancer carrying the exact mutant p53. Thus, novel antibodies targeting mutant p53, but not the wild-type isoform, should be pursued in preclincial and clinical studies.

Antioxidant Molecules from Plant Waste
The fruit, vegetable, legume, and cereal industries generate many wastes, representing an environmental pollution problem. However, these wastes are a rich source of antioxidant molecules such as terpenes, phenolic compounds, phytosterols, and bioactive peptides with potential applications mainly in the food and pharmaceutical industries, and they exhibit multiple biological properties including antidiabetic, anti-obesity, antihypertensive, anticancer, and antibacterial properties. The aforementioned has increased studies on the recovery of antioxidant compounds using green technologies to value plant waste, since they represent more efficient and sustainable processes. In this review, the main antioxidant molecules from plants are briefly described and the advantages and disadvantages of the use of conventional and green extraction technologies used for the recovery and optimization of the yield of antioxidant naturals are detailed; finally, recent studies on biological properties of antioxidant molecules extracted from plant waste are presented here.

Endodontic materials have significantly improved dental treatment techniques in several aspects as they can be used for vital pulp treatments, as temporary root canal medication, in definitive fillings, in apical surgeries, and for regenerative procedures. Calcium silicate-based cement is a class of dental material that is used in Endodontics in direct contact with the dental structures, connective tissue, and bone. Because the material interacts with biological tissues and stimulates biomineralization processes, its properties are of major importance. The main challenge in endodontic treatments is the elimination of biofilms that are present in the root canal system anatomical complexities, as it remains even after chemical-mechanical preparation and disinfection procedures. Thus, an additional challenge for these biomaterials is to exert antimicrobial activity while maintaining their biological properties in parallel.
