The traffic-engineering methods of planning based on the predict-and-provide principle have self-enforcing effects of induced traffic and an unhealthy environment for humans as well as for the planet. Regardless of the huge negative effects and the recognition that a paradigm shift is emerging as the sum of methods and achievements developed by the community of academics, experts, practitioners, policymakers, and urban communities. Findings can have practical, effective implications as the determinants of a new transport policy paradigm that shows the way out of the trap of path dependency. The originality of the approach lies in having expanded and applied the concept of anomalies of the theory to the adverse effects of technologies and the mismatch between people and the modern urban environment.

Mitochondria, as the main site of cellular energy metabolism and the generation of oxygen free radicals, are the key switch for mitochondria-mediated endogenous apoptosis. Ca2+ is not only an important messenger for cell proliferation, but it is also an indispensable signal for cell death. Ca2+ participates in and plays a crucial role in the energy metabolism, physiology, and pathology of mitochondria. Mitochondria control the uptake and release of Ca2+ through channels/transporters, such as the mitochondrial calcium uniporter (MCU), and influence the concentration of Ca2+ in both mitochondria and cytoplasm, thereby regulating cellular Ca2+ homeostasis. Mitochondrial Ca2+ transport-related processes are involved in important biological processes of tumor cells including proliferation, metabolism, and apoptosis.

Hepatitis C Virus
HCV leads to chronic infection in many patients that may progress to liver cirrhosis and hepatocellular carcinoma (HCC). The interferon (IFN) response is a critical component of the antiviral innate immune response against HCV infection. IFN signaling promotes the expression of many factors that can block the viral replication cycle. These IFN-induced antiviral factors can act at every level of HCV infection by decreasing viral entry, replication, transcription, translation, packaging and release. However, the antiviral state can generate significant collateral damage to the cell, requiring very tight control over the magnitude and duration of the IFN response. This is partially achieved through IFN-mediated negative self-regulation that helps in the termination of the IFN response and the return to homeostasis. However, these negative regulatory mechanisms can be hijacked by HCV to increase viral replication and promote productive infections.

Cancer initiation and progression is an accumulation of genetic and epigenetic modifications. DNA methylation is a common epigenetic modification that regulates gene expression, and aberrant DNA methylation patterns are considered a hallmark of cancer. The human diet is a source of micronutrients, bioactive molecules, and mycotoxins that have the ability to alter DNA methylation patterns and are thus a contributing factor for both the prevention and onset of cancer. In this review, we summarize the literature on dietary micronutrients, bioactive compounds, and food-borne mycotoxins that affect DNA methylation patterns and identify their potential in the onset and treatment of cancer.

Oral squamous cell carcinoma (OSCC) is an invasive epithelial neoplasm that is influenced by various risk factors, with a low survival rate and an increasing death rate. In the past few years, with the verification of the close relationship between different types of cancers and the microbiome, research has focused on the compositional changes of oral bacteria and their role in OSCC. Generally, oral bacteria can participate in OSCC development by promoting cell proliferation and angiogenesis, influencing normal apoptosis, facilitating invasion and metastasis, and assisting cancer stem cells. The study findings on the association between oral bacteria and OSCC may provide new insight into methods for early diagnosis and treatment development.

Hepatitis B virus (HBV) infection is the major risk factor for hepatocellular carcinoma (HCC). Understanding the unique features for HBV-induced HCC can shed new light on the unmet needs in its early diagnosis and effective therapy. During decades of chronic hepatitis B, hepatocytes undergoing repeated damage and regeneration accumulate genetic changes predisposing to HCC development. In addition to traditional mutations in viral and cellular oncogenes, HBV integration into the cell chromosomes is an alternative genetic change contributing to hepatocarcinogenesis. A striking male dominance in HBV-related HCC further highlights an interaction between androgen sex hormone and viral factors, which contributes to the gender difference via stimulating viral replication and activation of oncogenes preferentially in male patients. Meanwhile, a novel circulating tumor biomarker generated by HBV integration shows great potential for the early diagnosis of HCC. These unique HBV-induced hepatocarcinogenic mechanisms provide new insights for the future development of superior diagnosis and treatment strategies.

High Asia, the high-altitude habitat of the snow leopard (Panthera uncia), covers the mountainous regions of Asia’s twelve countries, including Afghanistan, Bhutan, China, India, Kazakhstan, Kyrgyzstan, Mongolia, Nepal, Pakistan, Russia, Tajikistan, and Uzbekistan. The transboundary landscape occupied by snow leopards covers an area of about 3.0247 million square kilometers. The snow leopard is an umbrella species from the high-altitude transboundary Asian regions. It is a threatened carnivore of an elusive and secretive nature.
