Biological Activities of Rhamnan Sulfate
Monostroma nitidum is a green single-cell layered algae that grows on the southwest coast of Japan. It is often used for salad ingredients, boiled tsukudani, soups, etc., due to its health benefits. M. nitidum is composed of many cell aggregates, and the various substances that fill the intercellular space are dietary fibers, vitamins, and minerals. Rhamnan sulfate (RS), a sulfated polysaccharide, is main the component of the fiber extracted from M. nitidum. Recently, some biological properties of RS have been demonstrated by in vitro and in vivo studies that probably protect human subjects from viruses and ameliorate vascular dysfunction caused by metabolic disorders, especially lifestyle-related diseases.

The endoplasmic reticulum (ER) of mammalian cells is the central organelle for the maturation and folding of transmembrane proteins and for proteins destined to be secreted into the extracellular space. The proper folding of target proteins is achieved and supervised by a complex endogenous chaperone machinery. BiP, a member of the Hsp70 protein family, is the central chaperone in the ER. The chaperoning activity of BiP is assisted by ER-resident DnaJ (ERdj) proteins due to their ability to stimulate the low, intrinsic ATPase activity of BiP. Besides their co-chaperoning activity, ERdj proteins also regulate and tightly control the translation, translocation, and degradation of proteins. Three ERdj co-chaperones, ERdj1, ERdj2, and ERdj6, are functionally involved in the control of translation and translocation of ER target proteins.

The BlackDog is a pocket-sized, self-contained computer with a built-in biometric fingerprint reader which was developed in 2005 by Realm Systems, which is plugged into and powered by the USB port of a host computer using its peripheral devices for input and output. It is a mobile personal server which allows a user to use Linux, ones applications and data on any computer with a USB port. The host machine's monitor, keyboard, mouse, and Internet connection are used by the BlackDog for the duration of the session. As the system is self-contained and isolated from the host, requiring no additional installation, it is possible to make use of untrusted computers, yet using a secure system. Various hardware iterations exist, and the original developer Realm Systems closed down in 2007, being picked up by the successor Inaura, Inc.

Wire Arc Additive Manufacturing for Aluminum-Lithium Alloys
Out of all the metal additive manufacturing (AM) techniques, the directed energy deposition (DED) technique, and particularly the wire-based one, are of great interest due to their rapid production. In addition, they are recognized as being the fastest technique capable of producing fully functional structural parts, near-net-shape products with complex geometry and almost unlimited size. There are several wire-based systems, such as plasma arc welding and laser melting deposition, depending on the heat source. The main drawback is the lack of commercially available wire; for instance, the absence of high-strength aluminum alloy wires. Therefore, this entry covers conventional and innovative processes of wire production and includes a summary of the Al-Cu-Li alloys with the most industrial interest in order to foment and promote the selection of the most suitable wire compositions. The role of each alloying element is key for specific wire design in WAAM; this entry describes the role of each element (typically strengthening by age hardening, solid solution and grain size reduction) with special attention to lithium. At the same time, the defects in the WAAM part limit its applicability. For this reason, all the defects related to the WAAM process, together with those related to the chemical composition of the alloy, are mentioned. Finally, future developments are summarized, encompassing the most suitable techniques for Al-Cu-Li alloys, such as PMC (pulse multicontrol) and CMT (cold metal transfer).

Non-celiac Gluten Sensitivity
Gluten is a protein contained in wheat, barley and rye and will be found in a variety of everyday food items. It allows doughs to be more elastic and pliable, this is why gluten-containing grains have been used in breads and other baked goods for many years. It can be difficult to actually know if you have intolerance to gluten as there may be little to no symptoms.  The study aims to discover if there are any correlations between gastric distress and severity of distress post gluten consumption and the variants in rs2305764 (MYO9B). The gene MYO9B encodes a member of the myosin family of actin-based molecular motor heavy chain proteins, polymorphisms in the genes are associated with celiac disease, ulcerative colitis and general autoimmunity. 89 female (24-43y/o) with no diagnosis of celiac disease were tested under a custom Illumina array in conjunction with Muhdo Health Ltd, this array analyses 1000 SNPs, one of which is rs2305764 that was read in the forward/forward orientation. Of these 21 had CC, 28 had CT and 40 had TT, there were no “no calls”.  The results show that 9.5% of those with CC (n=21) had some form of gastric distress post gluten intake, 32.1% of those with CT (n=28) had some form of gastric distress post gluten intake and 57.4% of those with TT (n=40) had some form of gastric distress post gluten intake. The average for all participants was 38.2%. On gastric distress the average score for the CC group was 2.5 (mild – somewhat uncomfortable), the average score for the CT group was 2.7 (mild – somewhat uncomfortable) and for the TT group was 5.2 (discomfort, some pain and distress).

IL-10, an immunosuppressive cytokine, is considered an important anti-inflammatory modulator of glial activation, preventing inflammation-mediated neuronal degeneration under pathological conditions. In this narrative review, we summarize recent insights about the role of IL-10 in the neurodegeneration associated with neuroinflammation, in diseases such as Multiple Sclerosis, Traumatic Brain Injury, Amyotrophic lateral sclerosis, Alzheimer's Disease, and Parkinson's Disease, focusing on the contribution of this cytokine not only in terms of protective action, but also as possibly responsible for clinical worsening. The knowledge of this double face of the same coin, regarding the biological role of the IL-10, could aid the development of targeted therapies useful for limiting neurodegenerative processes.

The tumor microenvironment (TME) has become the focus of interest in cancer research and treatment. It includes the extracellular matrix (ECM) and ECM-modifying enzymes that are secreted by cancer and neighboring cells. The ECM serves both to anchor the tumor cells embedded in it and as a means of communication between the various cellular and non-cellular components of the TME. The cells of the TME modify their surrounding cancer-characteristic ECM. This in turn provides feedback to them via cellular receptors, thereby regulating, together with cytokines and exosomes, differentiation processes as well as tumor progression and spread. Matrix remodeling is accomplished by altering the repertoire of ECM components and by biophysical changes in stiffness and tension caused by ECM-crosslinking and ECM-degrading enzymes, in particular matrix metalloproteinases (MMPs). These can degrade ECM barriers or, by partial proteolysis, release soluble ECM fragments called matrikines, which influence cells inside and outside the TME.

Galectin-3 is often secreted into biological fluids, like serum and urine, thus used as a biomarker. Tissue Biopsy-Based MSI Testing
Colorectal cancer (CRC) is the third most commonly-diagnosed cancer in the world and ranked second for cancer-related mortality in humans. Microsatellite instability (MSI) is an indicator for Lynch syndrome (LS), an inherited cancer predisposition, and a prognostic marker which predicts the response to immunotherapy. A recent trend in immunotherapy has transformed cancer treatment to provide medical alternatives that have not existed before. It is believed that MSI-high (MSI-H) CRC patients would benefit from immunotherapy due to their increased immune infiltration and higher neo-antigenic loads. MSI testing such as immunohistochemistry (IHC) and PCR MSI assay has historically been a tissue-based procedure that involves the testing of adequate tissue with a high concentration of cancer cells, in addition to the requirement for paired normal tissues. The invasive nature and specific prerequisite of such tests might hinder its application when surgery is not an option or when the tissues are insufficient. The application of next-generation sequencing, which is highly sensitive, in combination with liquid biopsy, therefore, presents an interesting possibility worth exploring. This review aimed to discuss the current body of evidence supporting the potential of liquid biopsy as a tool for MSI testing in CRC.
