Biological Control of Fall Armyworm
The fall armyworm (FAW), Spodoptera frugiperda, is one of the most important invasive pests worldwide, resulting in considerable losses in host crops. FAW comprises two genetic strains, such as the “rice strain”, which prefers rice and other grass species, and the “maize strain”, which feeds upon maize and sorghum. Potential control measures are generally more applicable to the farmers who lack financial assets to buy chemical insecticides or costly pure seeds. The adverse effects of pesticides on the ecosystem and human’s health and the development of resistance to insect pests have exaggerated efforts to find an alternative strategy that is cost-effective, low-risk and target-specific. Therefore, biological control is widely considered as one of the most important options for insect pest management.

Security Challenges and Malware Attacks in the IoT
Malware is a major security threat to the IoT, and detecting unknown malware is one of the key challenges for two reasons. First, the limitations of IoT devices, such as their low power retention capability and low computational processing capability, represent a significant challenge when aiming to apply security solutions. Second, introducing new ways to connect networks, such as cloud services, opens the door to many security attacks, such as malware attacks. Furthermore, connecting new devices that were not part of traditional networks via these new connection methods, such as smart sensors, makes applying security measures more complex. For these reasons, traditional malware detection mechanisms are not suitable for the IoT environment.

Phosphorylation Signals Downstream of Dopamine Receptors in NAc
Dopamine regulates emotional behaviors, including rewarding and aversive behaviors, through the mesolimbic dopaminergic pathway, which projects dopamine neurons from the ventral tegmental area to the nucleus accumbens (NAc). Protein phosphorylation is critical for intracellular signaling pathways and physiological functions, which are regulated by neurotransmitters in the brain. Previous studies have demonstrated that dopamine stimulated the phosphorylation of intracellular substrates, such as receptors, ion channels, and transcription factors, to regulate neuronal excitability and synaptic plasticity through dopamine receptors. Recent advances in proteomics techniques have clarified the mechanisms through which dopamine controls rewarding and aversive behaviors through signal pathways in the NAc.

Aneurysm prevention in Kawasaki Disease
 A form of systemic vasculitis that affects mostly small and medium-sized vessels, Kawasaki disease (KD) is most commonly found in children under the age of 5 years old. Though its etiology is unknown, KD has been the most frequent acquired heart disease in developing countries. The most severe complications of KD are coronary artery lesions (CAL), including dilation, fistula, aneurysm, arterial remodeling, stenosis, and occlusion. Aneurysm formation has been observed in 20-25% of KD patients that do not receive intravenous immunoglobulin (IVIG) treatment, and in 3-5% that do receive it. Coronary artery dilation has been found in about 30% of KD patients in the acute stage, although mostly in the transient form. Diminishing the occurrence and regression of aneurysm is a vital part of treating KD. This review is focuing on coronary artery aneurysm prevention in KD.

OTOF-Associated Hearing Impairment
Otoferlin, an essential synaptic protein in the auditory sensory inner hair cells, is encoded by the gene OTOF. Biallelic variants in OTOF are associated with autosomal recessive auditory neuropathy or synaptopathy (DFNB9). Since its discovery in 1996, roughly 220 causally-associated variants have been uncovered. The prevalence of OTOF-associated hearing loss varies according to population. A higher molecular genetic diagnostic yield has been reported in patients with prelingual auditory synaptopathy, making this very specific clinical feature a useful criterion for OTOF diagnostic testing. Apart from this clinical hallmark, genotype-phenotype correlations are rather complex and include a variant that causes temperature-sensitive auditory neuropathy (p.Ile515Thr) and three variants that have so far been associated with progressive hearing impairment. Several founder variants have been reported in the Asian and European populations. A complete landscape of genomic variation at the DFNB9 locus remains to be determined and improvements in genomics technologies may eventually uncover novel insights into possible missing variants. A complete understanding of OTOF variants is essential for the success of current and future therapies.

Gene Therapy for Cholestasis
Cholestatic diseases can be caused by the dysfunction of transporters involved in hepatobiliary circulation. Although pharmacological treatments constitute the current standard of care for these diseases, none are curative, with liver transplantation being the only long-term solution for severe cholestasis, albeit with many disadvantages. Liver-directed gene therapy has shown promising results in clinical trials for genetic diseases, and it could constitute a potential new therapeutic approach for cholestatic diseases. Gene therapy has emerged as a promising approach to achieve safe, stable, and efficient long-term correction for a wide range of genetic diseases, including monogenic liver disorders, for which liver transplantation remains the only cure, as well as acquired liver diseases.

Screening
Screening, in medicine, is a strategy used to look for as-yet-unrecognised conditions or risk markers. This testing can be applied to individuals or to a whole population. The people tested may not exhibit any signs or symptoms of a disease, or they might exhibit only one or two symptoms, which by themselves do not indicate a definitive diagnosis. Screening interventions are designed to identify conditions which could at some future point turn into disease, thus enabling earlier intervention and management in the hope to reduce mortality and suffering from a disease. Although screening may lead to an earlier diagnosis, not all screening tests have been shown to benefit the person being screened; overdiagnosis, misdiagnosis, and creating a false sense of security are some potential adverse effects of screening. Additionally, some screening tests can be inappropriately overused. For these reasons, a test used in a screening program, especially for a disease with low incidence, must have good sensitivity in addition to acceptable specificity. Several types of screening exist: universal screening involves screening of all individuals in a certain category (for example, all children of a certain age). Case finding involves screening a smaller group of people based on the presence of risk factors (for example, because a family member has been diagnosed with a hereditary disease). Screening interventions are not designed to be diagnostic, and often have significant rates of both false positive and false negative results. Frequently updated recommendations for screening are provided by the independent panel of experts, the United States Preventive Services Task Force.

P53 DNA Binding Cooperativity Mutations
p53 is a tumor suppressor that is mutated in half of all cancers. The high clinical relevance has made p53 a model transcription factor for delineating general mechanisms of transcriptional regulation. p53 forms tetramers that bind DNA in a highly cooperative manner. The DNA binding cooperativity of p53 has been studied by structural and molecular biologists as well as clinical oncologists. These experiments have revealed the structural basis for cooperative DNA binding and its impact on sequence specificity and target gene spectrum. Cooperativity was found to be critical for the control of p53-mediated cell fate decisions and tumor suppression. Importantly, an estimated number of 34,000 cancer patients per year world-wide have mutations of the amino acids mediating cooperativity, and knock-in mouse models have confirmed such mutations to be tumorigenic. While p53 cancer mutations are classically subdivided into “contact” and “structural” mutations, “cooperativity” mutations form a mechanistically distinct third class that affect the quaternary structure but leave DNA contacting residues and the three-dimensional folding of the DNA-binding domain intact. In this review we discuss the concept of DNA binding cooperativity and highlight the unique nature of cooperativity mutations and their clinical implications for cancer therapy.
