Cardiovascular diseases are one of the leading causes of global death in developing countries. More than 80% of cardiovascular disease-associated mortality is attributable to atherosclerosis, a chronic inflammatory disease of the vessel wall. During the development of atherosclerosis and other cardiovascular diseases, vascular smooth muscle cells (SMCs) continuously shift from a contractile state towards other phenotypes that differ substantially from differentiated SMCs.

Human Sirtuins
Sirtuins (SIRTs) are nicotinamide adenine dinucleotide-dependent histone deacetylases that in-corporate complex functions in the mechanisms of cell physiology. Mammals have seven distinct members of the SIRT family (SIRT1-7), which play an important role in a well-maintained net-work of metabolic pathways that control and adapt the cell to the environment, energy availabil-ity and cellular stress. Until recently, very few studies investigated the role of SIRTs in modulating viral infection and progeny. Recent studies have demonstrated that SIRT1 and SIRT2 are promis-ing antiviral targets because of their specific connection to numerous metabolic and regulatory processes affected during infection.

ClpP
The mitochondrial protease called ClpP plays a central role in mitochondrial protein quality control. ClpP agonists have emerged as a novel class of mitochondria-targeting drugs. Hyperactivating ClpP induces uncontrolled, but selective, degradation of ClpP substrates and disrupts mitochondrial functions, leading to growth inhibition of breast cancer cells, without adverse effect in non-malignant cells.

The Guanyl Radical
Guanyl radical or neutral guanine radical G(-H)• results from the loss of a H-atom or an electron/proton (e–/H+) couple from the guanine structures (G). Guanyl radical G(-H)• exists in two tautomeric forms and its role in single- and double-stranded oligonucleotides, in DNA and G-quadruplex, attracted considerable attention since directly connected to the damage of genetic material and its biological consequences. The emerging picture is still incomplete and extrapolation of its chemistry from nucleosides to more complex environment like DNA can be misleading.

Targeting CDK4/6 for Anticancer Therapy
Cyclin-dependent kinase 4/6 (CDK4/6) are key regulators of the cell cycle and are deemed as critical therapeutic targets of multiple cancers. Various approaches have been applied to silence CDK4/6 at different levels, i.e., CRISPR to knock out at the DNA level, siRNA to inhibit translation, and drugs that target the protein of interest. Here we summarize the current status in this field, highlighting the mechanisms of small molecular inhibitors treatment and drug resistance.

BRCA1 in Centrosome Regulation
Breast cancer gene 1 (BRCA1) is a tumor suppressor associated with hereditary breast and ovarian cancer and forms a heterodimer with BRCA1-associated RING domain protein 1 (BARD1). Centrosomes are the major microtubule-organizing centers in animal cells and are critical for the formation of a bipolar mitotic spindle. BRCA1 and BARD1 localize to the centrosome during the cell cycle, and the BRCA1/BARD1 dimer ubiquitinates centrosomal proteins to regulate centrosome function.

Adipose Tissue and FoxO1
Forkhead box O class proteins (FoxOs) are expressed nearly in all tissues and are involved in different functions such as energy metabolism, redox homeostasis and differentiation. The plasticity of FoxOs is demonstrated by post-translational modifications that determine diverse levels of transcriptional regulations also controlled by their subcellular localization. Among the different members of the FoxO family, we focused on FoxO1 in adipose tissue, where it is abundantly expressed and is involved in differentiation and transdifferentiation processes. The capability of FoxO1 to respond differently in dependence of adipose tissue subtype underlines the specific involvement of the transcription factor in energy metabolism and the “browning” process of adipocytes. FoxO1 can localize to nuclear, cytoplasm, and mitochondrial compartments of adipocytes responding to different availability of nutrients and source of reactive oxygen species (ROS).
