The ultimate goal of liver tissue engineering (LTE)  is to restore partial or total function of the liver during liver failure. A fully functional liver is the ultimate aim of LTE, and functional liver tissue can be used for drug testing. Moreover, LTE has the potential to develop an extracorporeal liver support (ECLS) system performing the essential functions of the liver to reduce mortality or to bridge a patient to a liver transplant. With recent advances in the field of regenerative medicine, tissue engineering holds high potential to progress as an alternative or supplement to LT. A major advance in the LTE area was published by Chhabra and colleagues, reporting the development of a vascularized liver model to understand liver regeneration.

Critical Limb Ischemia
Critical limb ischemia (CLI) constitutes the most severe form of peripheral arterial disease (PAD), a prevalent manifestation of atherosclerosis which involves the blockade of major systemic arteries other than those of the cerebral and coronary circulation, more common in legs than in arms. Overall, CLI patients suffer from chronic ischemic rest pain, ulcers, or gangrene, as well as an increased risk of cardiovascular events. CLI has a huge impact on the patients’ quality of life, being associated with an increased risk of amputations (fingers, toes, or extremities) and, moreover, an increase in mortality rates. Currently, revascularization strategies (bypass grafting, angioplasty) remain the first option for CLI patients, although less than 45% of them are eligible for surgical intervention mainly due to associated comorbidities. Moreover, patients usually require amputation in the short-term. As an alternative to conventional treatments, therapeutic angiogenesis has arisen as a promising treatment for CLI patients, mainly those considered as “no-option”, due to the potential of this strategy to promote revascularization of ischemic tissues. Different approaches including angiogenic gene or cell-based therapies are currently under investigation.

Ovine Paratuberculosis Control
Ovine paratuberculosis due to Mycobacterium avium subsp. paratuberculosis (MAP) is no longer the serious agripolitical animal health issue that it was for many Australian rural communities a decade and a half ago. However, the disease continues to spread, with both OJD extension and funded research programs required to address remaining knowledge gaps, the challenging issues of promulgated misinformation, and the need for continual improvement of diagnostic and disease control tools, respectively. Improved regional and on-farm biosecurity, including the introduction of a risk-based trading system, may have contributed to changing attitudes to OJD control, although decreasing on-farm OJD prevalence is almost certainly attributable mostly to the uptake of persistent vaccination programs with Gudair. Although encouraging the ongoing use of vaccination and improved biosecurity when OJD mortalities disappear remains challenging, vaccination has provided a robust strategy for managing OJD on-farm and contributed significantly to both the health of Australian sheep and the lives of producers with affected properties. As vaccination offers a pathway to reduce the risk of MAP infection entering the human food chain from ruminant products, it should be more widely adopted globally.

Anticancer Properties of Carnosol
Cancer is characterized by unrestricted cell proliferation, inhibition of apoptosis, enhanced invasion and migration, and deregulation of signaling cascades. These properties lead to uncontrolled growth, enhanced survival, and the formation of tumors. Carnosol, a naturally occurring phyto-polyphenol (diterpene) found in rosemary, has been studied for its extensive antioxidant, anti-inflammatory, and anticancer effects. In cancer cells, carnosol has been demonstrated to inhibit cell proliferation and survival, reduce migration and invasion, and significantly enhance apoptosis. These anticancer effects of carnosol are mediated by the inhibition of several signaling molecules including extracellular signal-regulated kinase (ERK), p38, c-Jun N-terminal kinase (JNK), Akt, mechanistic target of rapamycin (mTOR) and cyclooxygenase-2 (COX-2). Additionally, carnosol prevents the nuclear translocation of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) and promotes apoptosis, as indicated by increased levels of cleaved caspase-3, -8, -9, increased levels of the pro-apoptotic marker Bcl-2-associated X (BAX), and reduced levels of the anti-apoptotic marker B-cell lymphoma 2 (Bcl-2).
