Ocular Cystinosis
Cystinosis is a rare autosomal recessive disease that affects about 1 in 100,000–200,000 people among the general population, characterized by high levels of cystine within the lysosomes in cells of certain types of tissues. The accumulation of this substance is caused by mutations in the CTNS gene which codes for cystinosin, the carrier that transports cystine out of the lysosome. Fetal Hemoglobin in Sickle Hemoglobinopathies
Fetal hemoglobin (HbF; α2γ2), encoded by two nearly identical γ-globin genes (HBG2, HBG1) that are part of the β-globin gene (HBB) cluster (11p15.4), comprises 70 to 90% of the hemolysate in newborns, falling to <1% after 12 months. HbF usually consists of 4 to 10% of total hemoglobin in adults of African descent with sickle cell anemia. Rarely, their HbF levels reach more than 30%. High HbF levels are sometimes a result of β-globin gene deletions or point mutations in the promoters of the HbF genes.

Mining Effects on the Karst
Karst develops on soluble rocks (limestone, dolomite, and evaporite). The infiltrating water with carbonic acid creates cavities (caves), fills them, and flows towards the mountain margin (karst water), where it emerges in springs. The infiltrated water constitutes a three-dimensional system whose surface is the karst water level, which undergoes fluctuation of various degrees and periods due to natural and artificial effects, at another time a one-way rise or subsidence. Since karst rocks drain water, neither a surface water network nor valleys develop (they are only formed at sites where the valley is inherited from the non-karstic cover or when the karst water level is situated at the valley floor). The dissolved material is transported into the karst with the infiltrating waters; therefore, surface karst features are closed. These are karren, dolines, ponors with blind valleys, and poljes. The material transported in the solution precipitates as freshwater limestone.

Morphometric Characterization of Bacteria Associated with Bacteremia
Among the leading causes of bacteremia are Escherichia coli, Klebsiella pneumoniae, and Staphylococcus aureus. E. coli and K. pneumoniae are increasingly exhibiting resistance to last-resort antibiotics, such as carbapenems. Rapid and accurate identification of these pathogens is critical for timely treatment and infection control. This paper aimed to develop a computer-aided bacterial morphometric technique for identifying and classifying wild-type E. coli, K. pneumoniae, and S. aureus in a field guide fashion. A 3D laser scanning confocal microscope was used to gather key parameters of each organism: length (L, µm), circular diameter (CD, µm), volume (V, µm3), surface area-to-cross-sectional area ratio (SA/CSA, unitless), surface uniformity ratio (Str), and surface texture ratio (Sdr). Microscope images and measurement results showed that S. aureus was spherical with the shortest length (1.08 µm) and smallest volume (0.52 µm3). E. coli and K. pneumoniae were rod-shaped with lengths >2.0 µm and volumes >1.0 µm3. Carbapenem-resistant (CR) strains exhibited larger volumes than their wild-type counterparts. Surface parameters further differentiated strains: wild-type E. coli had a greater surface texture or a less smooth surface (larger Sdr) than K. pneumoniae (lower Sdr) did. CR E. coli had more surface uniformity (lower Str) than CR K. pneumoniae did. A dichotomous key based on shape, circular diameter, volume, length, and surface characteristics was developed to classify the species using a series of paired, contrasting features. This morphometric analysis can aid researchers in quickly identifying bacteria, leading to faster diagnosis of life-threatening diseases and improved treatment decisions.

Recombinant Adeno-Associated Viral Vectors (rAAV)-Vector Elements
Inherited retinal dystrophies and optic neuropathies cause chronic disabling loss of visual function. The development of recombinant adeno-associated viral vectors (rAAV) gene therapies in all disease fields have been promising, but the translation to the clinic has been slow. The safety and efficacy profiles of rAAV are linked to the dose of applied vectors. DNA changes in the rAAV gene cassette affect potency, the expression pattern (cell-specificity), and the production yield. Here, we present a library of rAAV vectors and elements that provide a workflow to design novel vectors. We first performed a meta-analysis on recombinant rAAV elements in clinical trials (2007-2020) for ocular gene therapies. We analyzed 33 unique rAAV gene cassettes used in 57 ocular clinical trials. The rAAV gene therapy vectors used six unique capsid variants, 16 different promoters, and six unique polyadenylation sequences. Further, we compiled a list of promoters, enhancers, and other sequences used in current rAAV gene cassettes in preclinical studies. Then, we give an update on pro-viral plasmid backbones used to produce the gene therapy vectors, inverted terminal repeats, production yield, and rAAV safety considerations. Finally, we assess rAAV transgene and bioactivity assays applied to cells or organoids in vitro, explants ex vivo, and clinical studies.

Osteoclast Multinucleation
       Osteoclasts  are derived from hemopoietic progenitors of the monocyte-macrophage lineage. They differentiate upon exposure to macrophage colony stimulating factor (M-CSF) and receptor activator of NF-κB ligand (RANKL), which are presented by osteoblasts and osteocytesare. Multinucleation in a late phase of osteoclastogenesis is a hallmark of osteoclast maturation. The unique and dynamic multinucleation process not only increases cell size but causes functional alterations through reconstruction of the cytoskeleton, creating the actin ring and ruffled border that enable efficient bone resorption.  The process of osteoclast multinucleation is dynamic, complicated and finely controlled by multiple entangled factors. At the beginning of the 21st century, two master fusogens, DC-STAMP and OC-STAMP, had been identified that directly regulate osteoclast multinucleation.

LaGaO3-Based Solid Oxide Fuel Cell Electrolytes
Solid oxide fuel cells (SOFCs) are efficient electrochemical devices that allow for the direct conversion of fuels (their chemical energy) into electricity. Although conventional SOFCs based on YSZ electrolytes are widely used from laboratory to commercial scales, the development of alternative ion-conducting electrolytes is of great importance for improving SOFC performance at reduced operation temperatures. The basic information has been studied on representative family of oxygen-conducting electrolytes, such as doped lanthanum gallates (LaGaO3). Complex oxides based on LaGaO3 offer a convenient basis for the design of oxygen-conducting electrolytes that can be employed in intermediate-temperature solid oxide fuel cells. A rational combination of appropriate dopants incorporated at various sublattices of LaGaO3 allows superior transport properties to be achieved for co-doped derivatives (La1−xSrxGa1−yMgyO3−δ, LSGM).
