Thermostable O6-alkylguanine-DNA Alkyltransferases
The genome of living cells is continuously exposed to endogenous and exogenous attacks, and this is particularly amplified at high temperatures. Alkylating agents cause DNA damage, leading to mutations and cell death; for this reason, a class of enzymes known as alkylguanine-DNA-alkyltransferases (AGTs) protects the DNA from mutations caused by alkylating agents, in particular in the recognition and repair of alkylated guanines in O6-position. The peculiar irreversible self-alkylation reaction of these enzymes triggered numerous studies, especially on the human homologue, in order to identify effective inhibitors in the fight against cancer. In modern biotechnology, engineered variants of AGTs are developed to be used as protein tags for the attachment of chemical ligands. In the last decade, research on AGTs from (hyper)thermophilic sources proved useful as a model system to clarify numerous phenomena, also common for mesophilic enzymes. This review traces recent progress in this class of thermozymes, emphasizing their usefulness in basic research and their consequent advantages for in vivo and in vitro biotechnological applications.

School Design on Users’ Responses
This systematic review focused on the effect of the educational environment design on students’ and teachers’ performance, satisfaction, and wellbeing. Starting from a bulk of 1307 articles, a set of N = 68 empirical papers was selected and organized on the basis of four different content clusters, i.e., architectural building design and aesthetic features, indoor environmental features, classroom design, and school green spaces/outdoor spaces. From the analysis of research findings, the key role of pleasant, warm, and flexible learning environments emerged, for promoting both wellbeing and performance of users. More specifically, the presence of charming colors and pictures, ergonomic furniture, and adequate acoustic, thermal comfort, ventilation, and natural lighting have emerged as important features that school designers should care for. Furthermore, an integration of both indoor and outdoor learning situations showed to be effective for improving students’ learning and wellbeing.

CMOS Sensors
Depleted Complementary Metal-Oxide-Semiconductor (CMOS) sensors are emerging as one of the main candidate technologies for future tracking detectors in high luminosity colliders. Their capability of integrating the sensing diode into the CMOS wafer hosting the front-end electronics allows for reduced noise and higher signal sensitivity, due to the direct collection of the sensor signal by the readout electronics. They are suitable for high radiation environments due to the possibility of applying high depletion voltage and the availability of relatively high resistivity substrates. The use of a CMOS commercial fabrication process leads to their cost reduction and allows faster construction of large area detectors. In this contribution, a general perspective of the state of the art of CMOS detectors for High Energy Physics experiments is given. The main developments carried out with regard to these devices in the framework of the CERN RD50 collaboration are summarized.

Bisphenol A-Induced Male Infertility
Bisphenol A (BPA) is a well-known endocrine disruptor present in epoxy resins and polycarbonate plastics, which negatively disturbs the male reproductive system affecting male fertility. In vivo studies showed that BPA exposure has deleterious effects on spermatogenesis by disturbing the hypothalamic-pituitary-gonadal axis and inducing oxidative stress in the testis. This compound seems to disrupt hormone signalling even at low concentrations, modifying the levels of inhibin B, oestradiol, and testosterone. The adverse effects on seminal parameters are mainly supported by studies based on urinary BPA concentration, showing a negative association between BPA levels and sperm concentration, motility, normal morphology and sperm DNA damage.

Burnout
Maslach et al. initially conceptualized burnout as a psychological state encompassing exhaustion, depersonalization (or cynicism), and a reduced sense of professional efficacy. However, Schaufeli et al. recently proposed an improved definition of burnout, as a syndrome reflecting: “a work-related state of exhaustion that occurs among employees, which is characterized by extreme tiredness, reduced ability to regulate cognitive and emotional processes, and mental distancing” (p. 4). This new definition excludes the professional efficacy component, which has been empirically found to be distinct from burnout. More generally, burnout seems to be primarily characterized by a state of work-related exhaustion and is generally acknowledged by researchers, clinicians, and the public as a pervasive occupational difficulty. However, and despite this widespread recognition, longstanding debates remain within the scientific community regarding the appropriateness of classifying burnout as its own pathological entity.

Special Prosecutor
In the United States, a special counsel (formerly called special prosecutor or independent counsel) is a lawyer appointed to investigate, and potentially prosecute, a particular case of suspected wrongdoing for which a conflict of interest exists for the usual prosecuting authority. Other jurisdictions have similar systems. For example, the investigation of an allegation against a sitting president or attorney general might be handled by a special prosecutor rather than by an ordinary prosecutor who would otherwise be in the position of investigating their own superior. Investigations into other persons connected to the government but not in a position of direct authority over the prosecutor, such as cabinet secretaries or election campaigns, have also been handled by special prosecutors. While the most prominent special prosecutors have been those appointed since the 1870s to investigate presidents and those connected to them, the term can also be used to refer to any prosecutor appointed to avoid a conflict of interest or appearance thereof. The concept originates in state law: "state courts have traditionally appointed special prosecutors when the regular government attorney was disqualified from a case, whether for incapacitation or interest." Because district attorneys' offices work closely with police, some activists argue that cases of police misconduct at the state and local level should be handled by special prosecutors.

Physical Exercise and the Hallmarks of Breast Cancer
Growing evidence suggests that, among the different molecular/cellular pathophysiological mechanisms associated with cancer, there are 14 hallmarks that play a major role, including: (i) sustaining proliferative signaling, (ii) evading growth suppressors, (iii) activating invasion and metastasis, (iv) enabling replicative immortality, (v) inducing angiogenesis, (vi) resisting cell death, (vii) reprogramming energy metabolism, (viii) evading immune destruction, (ix) genome instability and mutations, (x) tumor-promoting inflammation, (xi) unlocking phenotypic plasticity, (xii) nonmutational epigenetic reprogramming, (xiii) polymorphic microbiomes, and (xiv) senescent cells. These hallmarks are also associated with the development of breast cancer, which represents the most prevalent tumor type in the world. For the first time, the effects of physical activity/exercise on these hallmarks are described. In summary, an active lifestyle, and particularly regular physical exercise, provides beneficial effects on all major hallmarks associated with breast cancer, and might therefore help to counteract the progression of the disease or its associated burden.

Furthermore, these plant derivatives are widely interested in human and animal health, including potent antitumor, antifungal, anti-inflammatory, and bactericidal activity. Given this diversity, different methodologies were needed to optimize the extraction, purification, and characterization of each class of biomolecules. In addition, these plant products can still be used in the synthesis of nanomaterials to reduce the undesirable effects of conventional synthesis routes based on hazardous/toxic chemical reagents and associate the properties of nanomaterials with those present in extracts and essential oils.

Real Driving Emissions
Air pollution caused by vehicle emissions has raised serious public health concerns. Vehicle emissions generally depend on many factors, such as the nature of the vehicle, driving style, traffic conditions, emission control technologies, and operational conditions. Concerns about the certification cycles used by various regulatory authorities are growing due to the difference in emission during certification procedure and Real Driving Emissions (RDE). Under laboratory conditions, certification tests are performed in a ‘chassis dynamometer’ for light-duty vehicles (LDVs) and an ‘engine dynamometer’ for heavy-duty vehicles (HDVs). As a result, the test drive cycles used to measure the automotive emissions do not correctly reflect the vehicle’s real-world driving pattern. Consequently, the RDE regulation is being phased to reduce the disparity between type approval and vehicle’s real-world emissions. According to this review, different variables such as traffic signals, driving dynamics, congestions, altitude, ambient temperature, and so on have a major influence on actual driving pollution. Aside from that, cold-start and hot-start have been shown to affect on-road pollution. Contrary to common opinion, new technology such as start-stop systems boost automotive emissions rather than decreasing them owing to unfavourable conditions from the point of view of exhaust emissions and exhaust after-treatment systems. In addition, the driving dynamics are not represented in the current laboratory-based test procedures. As a result, it is critical to establish an on-road testing protocol to obtain a true representation of vehicular emissions and reduce emissions to a standard level. The incorporation of RDE clauses into certification procedures would have a positive impact on global air quality.

In Vitro Placental Models of Human Trophoblast
The complex process of placental implantation and development affects trophoblast progenitors and uterine cells through the regulation of transcription factors, cytokines, adhesion receptors and their ligands. Differentiation of trophoblast precursors in the trophectoderm of early ontogenesis, caused by the transcription factors, such as CDX2, TEAD4, Eomes and GATA3, leads to the formation of cytotrophoblast and syncytiotrophoblast populations. The molecular mechanisms involved in placental formation inside the human body along with the specification and differentiation of trophoblast cell lines are, mostly due to the lack of suitable cell models, not sufficiently elucidated. This research is an evaluation of current technologies, which are used to study the behavior of human trophoblasts and other placental cells, as well as their ability to represent physiological conditions both in vivo and in vitro. An in vitro 3D model with a characteristic phenotype is of great benefit for the study of placental physiology. At the same time, it provides great support for future modeling of placental disease.
