Treatment Combinations with DNA Vaccines
Metastatic castration-resistant prostate cancer (mCRPC) is a challenging disease to treat, with poor outcomes for patients. One antitumor vaccine, sipuleucel-T, has been approved as a treatment for mCRPC. DNA vaccines are another form of immunotherapy under investigation. DNA immunizations elicit antigen-specific T cells that cause tumor cell lysis, which should translate to meaningful clinical responses. They are easily amenable to design alterations, scalable for large-scale manufacturing, and thermo-stable for easy transport and distribution. Hence, they offer advantages over other vaccine formulations. However, clinical trials with DNA vaccines as a monotherapy have shown only modest clinical effects against tumors. Standard therapies for CRPC including androgen-targeted therapies, radiation therapy and chemotherapy all have immunomodulatory effects, which combined with immunotherapies such as DNA vaccines, could potentially improve treatment. In addition, many investigational drugs are being developed which can augment antitumor immunity, and together with DNA vaccines can further enhance antitumor responses in preclinical models.

Retroviruses selectively package two copies of their RNA genomes from a cellular milieu that includes a substantial excess of host and non-genomic viral RNAs. Present understanding of the structural determinants and mechanism of retroviral genome packaging has been derived from combinations of genetic experiments, phylogenetic analyses, nucleotide accessibility mapping, in silico RNA structure predictions, and biophysical studies. Genetic experiments provided early clues regarding the protein and RNA elements required for packaging, and nucleotide accessibility mapping experiments provided insights into the secondary structures of functionally important elements in the genome. Three-dimensional structural determinants of packaging were primarily derived by nuclear magnetic resonance (NMR) spectroscopy. A key advantage of NMR, relative to other methods for determining biomolecular structure (such as X-ray crystallography), is that it is well suited for studies of conformationally dynamic and heterogeneous systems—a hallmark of the retrovirus packaging machinery.

Transfer Learning in Breast Cancer
Transfer learning is a machine learning approach that reuses a learning method developed for a task as the starting point for a model on a target task. The goal of transfer learning is to improve performance of target learners by transferring the knowledge contained in other (but related) source domains. As a result, the need for large numbers of target-domain data is lowered for constructing target learners. Due to this immense property, transfer learning techniques are frequently used in ultrasound breast cancer image analyses. In this study, we focus on transfer learning methods applied on ultrasound breast image classification and detection from the perspective of transfer learning approaches, pre-processing, pre-training models, and convolutional neural network (CNN) models. Finally, comparison of different works is carried out, and challenges—as well as outlooks—are discussed.

Site-directed spin labeling (SDSL) is a molecular biology technique in which paramagnetic spin labels are incorporated into the specific site of bio-macromolecules for investigation of structure and dynamic properties using electron paramagnetic resonance (EPR) spectroscopy. In SDSL, all native nondisulfide bonded cysteines are eliminated by changing them with another amino acid such as serine or an alanine. A unique cysteine residue is then incorporated into a recombinant protein using site-directed mutagenesis technique, and further reacted with sulfhydryl-specific nitroxide reagent to covalently generate a stable spin label side-chain. The development of SDSL approaches extended the application of EPR to almost any biological systems. Site-directed spin labeling  (SDSL) combined with EPR spectroscopy can provide structural dynamics of spin label side-chain, solvent accessibility, solvent polarity, and intra- or intermolecular distances between two spin labels of macromolecules.

Acquisition of Brassica Yellows Virus
Brassica yellows virus (BrYV) is a tentative species of the genus Polerovirus, which occurs widely, and mostly damages Brassicaceae plants in East Asia. Because BrYV cannot be transmitted mechanically, an insect-based transmission method is required for further virus research. Here, a reliable and unrestricted method is described, in which non-viruliferous aphids (Myzus persicae) acquired BrYV from transgenic Arabidopsis thaliana, harboring the full-length viral genome germinated from seeds and its frozen leaves. The aphids then transmitted the virus to healthy plants. There was no significant difference in acquisition rates between fresh and frozen infected leaves, although the transmission rate from frozen infected leaves was lower compared to fresh infected leaves. This simple novel method may be used to preserve viral inocula, evaluate host varietal resistance to BrYV, and investigate interactions among BrYV, aphids, and hosts.

Enumeration of Brettanomyces in Wine Using Impedance
Brettanomyces bruxellensis is a wine spoilage concern in wineries around the world. In order to maintain wine quality during storage and ageing, it is imperative to control and monitor this yeast. Being a fastidious slow growing yeast, which requires 5 to 14 days of incubation for visible growth in agar plates, it is difficult to detect growth (colonies) by conventional agar plate count method. Yeast enumeration by impedance was investigated because previous research using other microorganisms has shown that it is potentially faster than plate counting. The relationship between plate counting and impedance detection times was investigated for Brettanomyces inoculated in red wine samples. A linear relationship between log plate count concentrations and impedance detection times was found. Incubation time was reduced from 120 h down to 0.9 and 57.7 h for samples with 6.7 × 10E7 and 1.8 × 10E2 cfu/mL, respectively, using the ‘indirect’ impedance method. The ‘indirect’ impedance method has the potential to be used by the wine industry to control and monitor the Brettanomyces numbers in wines.
