A Long-Lasting PARP1-Activation Mediates Signal-Induced Gene Expression
PolyADP-ribosylation is an evolutionary conserved, reversible post-translational modification of proteins. Numerous nuclear proteins act as substrates of the abundant nuclear polyADP-ribose polymerase 1 (PARP1). In this modification, negatively charged ADP-ribose chains constructed on chromatin-bound proteins, cause their repulsion from the negatively charged DNA. In accordance, polyADP-ribosylation is a post-translational modification of proteins that causes relaxation of the highly condensed structure of the chromatin. Histone H1, which is bound to the linker DNA, located between the nucleosomes, is a prominent substrate of PARP1.

A Mycobacteriophage-Based Vaccine Platform for SARS-CoV-2
Bacteriophage-based vaccines can generate a protective immune response by safely introducing foreign antigens displayed on, encapsidated within, or genetically encoded by phage. Here authors describe recombinants of mycobacteriophage Bxb1 (a phage infecting Mycobacterium smegmatis) that covalently display and express antigenic peptides of the SARS-CoV-2 Spike protein. Several of these vaccine candidates produced Spike-specific antibodies in immunized mice, but the responses were not neutralizing. This mycobacteriophage-based vaccine platform can likely be improved if delivery of larger antigens is achieved.

A Novel Innate Immune Regulatory Player: LncRNAs
Long non-coding RNAs (lncRNAs) represent crucial transcriptional and post-transcriptional gene regulators during antimicrobial responses in the host innate immune system. Studies have shown that lncRNAs are expressed in a highly tissue- and cell-specific- manner and are involved in the differentiation and function of innate immune cells, as well as inflammatory and antiviral processes, through versatile molecular mechanisms. These lncRNAs function via the interactions with DNA, RNA, or protein in either cis or trans pattern, relying on their specific sequences or their transcriptions and processing. The dysregulation of lncRNA function is associated with various human non-infectious diseases, such as inflammatory bowel disease, cardiovascular diseases, and diabetes mellitus. Here, we provide an overview of the regulation and mechanisms of lncRNA function in the development and differentiation of innate immune cells, and during the activation or repression of innate immune responses. These elucidations might be beneficial for the development of therapeutic strategies targeting inflammatory and innate immune-mediated diseases.

A Promiscuous Bacterial P450
CYP102A1 (BM3) is a catalytically self-sufficient flavocytochrome fusion protein isolated from Bacillus megaterium, which displays similar metabolic capabilities to many drug-metabolizing human P450 isoforms. BM3′s high catalytic efficiency, ease of production and malleable active site makes the enzyme a desirable tool in the production of small molecule metabolites, especially for compounds that exhibit drug-like chemical properties. The engineering of select key residues within the BM3 active site vastly expands the catalytic repertoire, generating variants which can perform a range of modifications. This provides an attractive alternative route to the production of valuable compounds that are often laborious to synthesize via traditional organic means.

A Telemedicine Approach to Covid-19
        COVID-19 is an RNA virus that has caused a pandemic leading to death and disability on a global scale. The virus gains entry into the cell using host protein angiotensin-converting enzyme-2 (ACE-2) receptor. The viral envelope fuses with the cell membrane through the endosomal pathway, allowing the viral particle to enter the host cell, where it can release its RNA and replicate. After an initial asymptomatic period, common presenting signs and symptoms of COVID-19 infection are non-specific and may include those of a common cold or influenza such as fever, dry cough, myalgia, headache and fatigue. Sore throat and nasal congestion may also be noted. Gastrointestinal and other symptoms may occur and loss of sense of taste and smell is a frequent early symptom that may distinguish COVID-19 from other respiratory infections. The virus is highly contagious and there is no cure. In order to avoid transmission, social distancing, mask wearing and reduced person-to-person interaction are key.  Definitive diagnosis is based on reverse-transcription PCR of nasopharyngeal and oropharyngeal swab samples. The clinical spectrum of the disease ranges from mild to moderate or severe. In order to minimize spread of COVID-19 to both patients and healthcare workers, virtual evaluation of suspected cases is being widely implemented and has necessitated an adjustment to the new medium by all participants. The use of virtual technology for real-time telemedicine assessment for COVID-19 provisional diagnosis and categorization of severity allows patients to receive an appropriate level of care without endangering others. We created a protocol for implementing a remote visit using a computer or mobile device. This enables patient assessement without direct contact to determine the need to visit a healthcare facility or alternatively, to be managed at home. If stable, the patient can isolate and be monitored so that should their condition worsen, they can be directed to in-person care at a hospital. Vigilance in checking on the patient's condition is crucial because those who develop severe COVID-19 may not experience dyspnea and other signs of rapid deterioration until about 5-8 days after symptom onset.
