Cutting-Edge: Preclinical and Clinical Development of Opdualag
Immune checkpoint inhibitors (ICIs) have revolutionized medical practice in oncology since the FDA approval of the first ICI 11 years ago. In light of this, Lymphocyte-Activation Gene 3 (LAG-3) is one of the most important next-generation immune checkpoint molecules, playing a similar role as Programmed cell Death protein 1 (PD-1) and Cytotoxic T-Lymphocyte Antigen 4 (CTLA-4). 19 LAG-3 targeting molecules are being evaluated at 108 clinical trials which are demonstrating positive results, including promising bispecific molecules targeting LAG-3 simultaneously with other ICIs. Recently, a new dual anti-PD-1 (Nivolumab) and anti-LAG-3 (Relatimab) treatment developed by Bristol Myers Squibb (Opdualag), was approved by the Food and Drug Administration (FDA) as the first LAG-3 blocking antibody combination for unresectable or metastatic melanoma. This novel immunotherapy combination more than doubled median progression-free survival (PFS) when compared to nivolumab monotherapy (10.1 months versus 4.6 months).

Global Utilizations of Cement Kiln Dust
A large quantity of cement kiln dust is produced daily, associated with ordinary Portland cement production worldwide. Most of these quantities are disposed of in landfills to meet the environmental requirement. Meanwhile, only a small proportion can be recycled in cement manufacturing. Recently, utilizing cement kiln dust (CKD) in many commercial applications has significantly increased due to the potential benefits of adding CKD, in different percentages, to the engineering properties of the prepared product. Moreover, utilizing CKD as a partial replacement for cement in industrial applications reduces the overconsumption of natural resources used for cement manufacturing and promotes sustainability.

Roadrunner (Supercomputer)
Roadrunner was a supercomputer built by IBM for the Los Alamos National Laboratory in New Mexico, USA. The US$100-million Roadrunner was designed for a peak performance of 1.7 petaflops. It achieved 1.026 petaflops on May 25, 2008, to become the world's first TOP500 LINPACK sustained 1.0 petaflops system. In November 2008, it reached a top performance of 1.456 petaFLOPS, retaining its top spot in the TOP500 list. It was also the fourth-most energy-efficient supercomputer in the world on the Supermicro Green500 list, with an operational rate of 444.94 megaflops per watt of power used. The hybrid Roadrunner design was then reused for several other energy efficient supercomputers. Roadrunner was decommissioned by Los Alamos on March 31, 2013. In its place, Los Alamos commissioned a supercomputer called Cielo, which was installed in 2010. Cielo was smaller and more energy efficient than Roadrunner, and cost $54 million.

MRCPsych (Member of the Royal College of Psychiatrists) is a postnominal qualification awarded to physicians who have completed the prescribed training requirements and membership examinations mandated by the Royal College of Psychiatrists. Candidates must be registered with the GMC or a recognized medical board in their own country and should have undergone three years of training. Although initially a two-part exam, the examination has undergone a radical change in the past few years, notably in terms of focus and structure. Although originally a two-part exam, the current examination is 3 parts, consisting of 2 written papers (Paper A and Paper B) and a clinical exam (CASC). Paper A focuses on Neuroscience, Pharmacology, Psychology and theory, while Paper B focuses on Current Clinical Practice & Evidence within General Adult and the various subspecialties of Psychiatry, Epidemiology, Statistics, Critical Appraisal and Psychotherapy The Royal college of psychiatrists has announced the change of format of the CASC exam changes for the exam in January 2018. The 4 linked stations in the morning session will now become 8 single stations with 4 minute to read and 7 minutes to complete the task. 6 of these will be patient management focus and the other 2 history taking. The other 8 single stations remain the same format 90 s to read and 7 minutes for the exam.

Mineralogy and Geochemistry of Ferromanganese Crusts
Late Pleistocene–Holocene rocks from the western part of Cocos-Nazca Spreading Centre (C-NSC) include ferromanganese crusts that elucidate the geochemistry and mineralogy of a deep-sea geological setting. Geochemical, mineralogical and petrological signatures indicate complex formation influenced by mild hydrothermal processes. These crusts consist mostly of mixed birnessite, todorokite-buserite, and Mn-(Fe) vernadite with traces of diagenetic manganates (asbolane), Fe-oxides and oxyhydroxides or hydrothermally associated and relatively pure Mn-oxyhydroxides (manganite). The average Mn/Fe ratio is 2.7, which suggests predominant mixed hydrogenous-early diagenetic crusts with hydrothermal influences. The mean concentrations of three prospective metals (Ni, Cu and Co) are low: 0.17, 0.08 and 0.025 wt %, respectively. The total content of ΣREY is also low, and ranges from 81 to 741 mg/kg (mean 339 mg/kg).

Foreshadow (Security Vulnerability)
Foreshadow (known as L1 Terminal Fault (L1TF) by Intel) is a vulnerability that affects modern microprocessors that was first discovered by two independent teams of researchers in January 2018, but was first disclosed to the public on 14 August 2018. The vulnerability is a speculative execution attack on Intel processors that may result in the disclosure of sensitive information stored in personal computers and third-party clouds. There are two versions: the first version (original/Foreshadow) (CVE-2018-3615) targets data from SGX enclaves; and the second version (next-generation/Foreshadow-NG) (CVE-2018-3620 and CVE-2018-3646) targets virtual machines (VMs), hypervisors (VMM), operating systems (OS) kernel memory, and System Management Mode (SMM) memory. A listing of affected Intel hardware has been posted. Foreshadow is similar to the Spectre security vulnerabilities discovered earlier to affect Intel and AMD chips, and the Meltdown vulnerability that also affected Intel. However, AMD products, according to AMD, are not affected by the Foreshadow security flaws. According to one expert, " lets malicious software break into secure areas that even the Spectre and Meltdown flaws couldn't crack". Nonetheless, one of the variants of Foreshadow goes beyond Intel chips with SGX technology, and affects "all [Intel] Core processors built over the last seven years". Foreshadow may be very difficult to exploit, and there seems to be no evidence to date (15 August 2018) of any serious hacking involving the Foreshadow vulnerabilities. Nevertheless, applying software patches may help alleviate some concern, although the balance between security and performance may be a worthy consideration. Companies performing cloud computing may see a significant decrease in their overall computing power; individuals, however, may not likely see any performance impact, according to researchers. The real fix, according to Intel, is by replacing today's processors. Intel further states, "These changes begin with our next-generation Intel Xeon Scalable processors (code-named Cascade Lake), as well as new client processors expected to launch later this year ." On 16 August 2018, researchers presented technical details of the Foreshadow security vulnerabilities in a seminar, and publication, entitled "Foreshadow: Extracting the Keys to the Intel SGX Kingdom with Transient Out-of-Order Execution" at a USENIX security conference.

Mechanism of NETosis
Neutrophil extracellular traps (NETs) are net-like structures released from neutrophils. NETs predominantly contain cell-free deoxyribonucleic acid (DNA) decorated with histones and neutrophil granule proteins. Numerous extrinsic and intrinsic stimuli can induce the formation of NETs such as pathogens, cytokines, immune complexes, microcrystals, antibodies, and other physiological stimuli. The mechanism of NETosis induction can either be ROS-dependent or independent based on the catalase producing activity of the pathogen. NADPH is the source of ROS production, which in turn depends on the upregulation of Ca2+ production in the cytoplasm. ROS-independent induction of NETosis is regulated through toll-like receptors (TLRs). Besides capturing and eliminating pathogens, NETs also aggravate the inflammatory response and thus act as a double-edged sword.
