Severe COVID-19 Lung Infection
Periodontal bacteria dissemination into the lower respiratory tract may create favorable conditions for severe COVID-19 lung infection. Once lung tissues are colonized, cells that survive persistent bacterial infection can undergo permanent damage and accelerated cellular senescence. Consequently, several morphological and functional features of senescent lung cells facilitate SARS-CoV-2 replication. The higher risk for severe SARS-CoV-2 infection, the virus that causes COVID-19, and death in older patients has generated the question whether basic aging mechanisms could be implicated in such susceptibility. Mounting evidence indicates that cellular senescence, a manifestation of aging at the cellular level, contributes to the development of age-related lung pathologies and facilitates respiratory infections. Apparently, a relationship between life-threatening COVID-19 lung infection and pre-existing periodontal disease seems improbable. However, periodontal pathogens can be inoculated during endotracheal intubation and/or aspirated into the lower respiratory tract.

Exosomes Derived from Tumor
Exosomes are lipid membrane-enclosed vesicles released by all cell types that act at the paracrine or endocrine level to favor cell differentiation, tissue homeostasis, organ remodeling, and immune regulation. Their biosynthesis begins with a cell membrane invagination which generates an early endosome that matures to a late endosome. By inward budding of the late endosome membrane, a multivesicular body (MVB) with intraluminal vesicles (ILVs) is generated. The fusion of MVBs with the plasma membrane releases ILVs into the extracellular space as exosomes, ranging in size from 30 to 100 nm in diameter. The bilipid exosome membrane is rich in cholesterol, ceramides, and phosphatidylserine and can be loaded with DNA, RNA, microRNAs, proteins, and lipids. It has been demonstrated that exosome secretion is a common mechanism used by the tumor to generate an immunosuppressive microenvironment that favors cancer development and progression, allowing tumor escape from immune control.

The Alter Retina
Alternative splicing of mRNA is an essential mechanism to regulate and increase the diversity of the transcriptome and proteome. Alternative splicing frequently occurs in a tissue- or time-specific manner, contributing to differential gene expression between cell types during development. Neural tissues present extremely complex splicing programs and display the highest number of alternative splicing events. As an extension of the central nervous system, the retina constitutes an excellent system to illustrate the high diversity of neural transcripts. The retina expresses retinal specific splicing factors and produces a large number of alternative transcripts, including exclusive tissue-specific exons, which require an exquisite regulation. In fact, a current challenge in the genetic diagnosis of inherited retinal diseases stems from the lack of information regarding alternative splicing of retinal genes, as a considerable percentage of mutations alter splicing or the relative production of alternative transcripts. Modulation of alternative splicing in the retina is also instrumental in the design of novel therapeutic approaches for retinal dystrophies, since it enables precision medicine for specific mutations.

Evolution of TFs and Enhancers
Adaptive immunity relies on the V(D)J DNA recombination of immunoglobulin (Ig) and T cell receptor (TCR) genes, which enables the recognition of highly diverse antigens and the elicitation of antigen-specific immune responses. This process is mediated by recombination-activating gene (Rag) 1 and Rag2 (Rag1/2), whose expression is strictly controlled in a cell type-specific manner; the expression of Rag1/2 genes represents a hallmark of lymphoid lineage commitment. Although Rag genes are known to be evolutionally conserved among jawed vertebrates, how Rag genes are regulated by lineage-specific transcription factors (TFs) and how their regulatory system evolved among vertebrates have not been fully elucidated. Here, we review the current body of knowledge concerning the cis-regulatory elements (CREs) of Rag genes and the evolution of the basic helix-loop-helix TF E protein regulating Rag gene CREs, as well as the evolution of the antagonist of this protein, the Id protein. This may help to understand how the adaptive immune system develops along with the evolution of responsible TFs and enhancers.

Business Process Re-Engineering to Digitalise Quality Control Checks
Business process re-engineering allows optimising processes within businesses. One way to do so is to use lean thinking to maximise customer value whilst minimising waste. Just-in-time is one of the core elements of lean production and is the material flow design upon which the lean production model is founded. For such a production model to succeed and achieve high efficiency and productivity levels, it needs to have a robust quality management system to accomplish the notion of ‘right first time’ and have quality controls at the source to prevent defects from entering the production system altogether. Defects occur when products are produced outside of agreed tolerances of the approved specification. Defects are a significant contributor to product waste and waste of resources, including raw materials, energy, water, and human resources. A reduction in waste generation helps to boost business performance by lowering environmental, economic and social impacts, i.e., the three pillars of sustainability. Optimised process monitoring helps identify defects in advance or as they happen in real time. A key function of the quality control (QC) process is to provide evidence that customer requirements (tolerances) of the product are met; therefore, measuring and controlling the values of the different variables that regulate the manufacturing processes is critical. However, collecting these values and handling the data manually tends to be slow, tedious, and prone to human error, as data can be misread, misplaced, or misrecorded easily. Furthermore, human judgement may not be consistent due to various factors including fatigue, mental or physical stress, as well as variability in heuristic and cognitive capabilities. Data capturing and analysing technology can be used instead to acquire such data and use it to control processes within acceptable parameters, ensuring optimum product flow and quality. Furthermore, as computing devices get smaller and cheaper, it is increasingly possible to tailor them to meet more needs on the factory floor, enabling machines to gather data, measure key performance indicators, and track operational efficiency. Data archiving is also necessary so that results can be reviewed later for auditing purposes. As cloud storage becomes safer, more affordable and with connection speeds that provide quick access, companies are increasingly moving away from paper records. This change introduces the challenge of reviewing and re-writing internal procedures to capture these new ways of data collection and reporting. It is important to know and prove who does what, when, how, and why for the purposes of traceability, accuracy, and consistency.

In some cases, CSC may be the source of all the tumor cells, the cause of the tumor’s resistance to chemotherapeutic agents, and the source of metastatic cells. Thus, a combination therapy targeting non-CSC tumor cells as well as specifically targeting CSCs holds the potential to be highly effective. Natural products (NPs) have been a historically rich source of biologically active compounds and are known for their ability to influence multiple signaling pathways simultaneously with negligible side effects. Natural flavonoids or potent derivatives are good candidates in exhibiting anti-CSC activity and targeting key functions required for CSC survival.

B-Cell Receptor Signaling Regulation and Aggressive B-Cell Lymphomas
The proliferation and survival signals emanating from the B-cell receptor (BCR) constitute a crucial aspect of mature lymphocyte’s life. Dysregulated BCR signaling is considered a potent contributor to tumor survival in different subtypes of B-cell non-Hodgkin lymphomas (B-NHLs). The emergence of BCR-associated kinases as rational therapeutic targets has led to the development and approval of several small molecule inhibitors targeting either Bruton’s tyrosine kinase (BTK), spleen tyrosine kinase (SYK), or phosphatidylinositol 3 kinase (PI3K), offering alternative treatment options to standard chemoimmunotherapy, and making some of these drugs valuable assets in the anti-lymphoma armamentarium. Despite their initial effectiveness, these precision medicine strategies are limited by primary resistance in aggressive B-cell lymphoma such as diffuse large B-cell lymphoma (DLBCL) and mantle cell lymphoma (MCL), especially in the case of first generation BTK inhibitors. In these patients, BCR-targeting drugs often fail to produce durable responses, and nearly all cases eventually progress with a dismal outcome, due to secondary resistance.
