Milk Exosomes
Milk contains various extracellular vesicles and non-vesicular structures: exosomes (with diameter 40-100 nm), vesicles of other size, fat globules (with diameter 4-6 mkm, containing milk fat globule membranes), and their aggregates. Due to the biocompatibility of milk exosomes, these vesicles have a wide potential as vehicles for oral delivery of therapeutically relevant molecules (drugs and therapeutic nucleic acids).

NK Cells in Cancer Therapy
       Cellular therapy has emerged as an attractive option for the treatment of cancer, and adoptive transfer of chimeric antigen receptor (CAR) expressing T cells has gained FDA approval in hematologic malignancy. However, limited efficacy was observed using CAR-T therapy in solid tumors. Natural killer (NK) cells are crucial for tumor surveillance and exhibit potent killing capacity of aberrant cells in an antigen-independent manner. Adoptive transfer of unmodified allogeneic or autologous NK cells has shown limited clinical benefit due to factors including low cell number, low cytotoxicity and failure to migrate to tumor sites. To address these problems, immortalized and autologous NK cells have been genetically engineered to express high affinity receptors (CD16), CARs directed against surface proteins (PD-L1, CD19, Her2, etc.) and endogenous cytokines (IL-2 and IL-15) that are crucial for NK cell survival and cytotoxicity, with positive outcomes reported by several groups both preclinically and clinically. With a multitude of NK cell-based therapies currently in clinic trials, it is likely they will play a crucial role in next-generation cell therapy-based treatment. In this review, we will highlight the recent advances and limitations of allogeneic, autologous and genetically enhanced NK cells used in adoptive cell therapy.

Role of Mitochondria in Neuroinflammation
Innate immune response is one of our primary defense against pathogens infection, although, if dysregulated, it represents the leading cause of chronic tissue inflammation. This dualism is even more present in the central nervous system, where neuroinflammation is both important for the activation of reparatory mechanisms and, at the same time, leads to the release of detrimental factors that induce neurons loss. Key players in modulating the neuroinflammatory response are mitochondria. They are responsible for a variety of cell mechanism that control tissue homeostasis, such as autophagy, apoptosis, energy production and also inflammation. Accordingly, it is widely recognized that mitochondria exert a pivotal role in the development of neurodegenerative diseases, since the neurodegenerative process is highly based on neuroinflammation and tissue damage. Interestingly, it has been suggested that neuroinflammation, and thus mitochondria (dys)function, have a fundamental role in neurodegenerative diseases and also in acute brain damage, such in ischemic stroke and epileptic seizures.

SARS-CoV-2 Main Protease
The main protease (Mpro) of the newly emerged severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was subjected to hyphenated pharmacophoric-based and structural-based virtual screenings using a library of microbial natural products (>24,000 compounds). Subsequent filtering of the resulted hits according to Lipinski’s rules was applied to select only the drug-like molecules. Top-scoring hits were further filtered out depending on their ability to show constant good binding affinities towards the molecular dynamic simulation (MDS)-derived enzyme’s conformers. Final MDS experiments were performed on the ligand-protein complexes to verify their binding modes and calculate their binding free energy. Consequently, a final selection of six compounds of microbial origin was proposed to possess high potential as anti-SARS-CoV-2 drug candidates. Our study provides insight into the role of the Mpro structural flexibility during interactions with the possible inhibitors and sheds light on the structure-based design of anti-coronavirus disease 2019 (COVID-19) therapeutics targeting SARS-CoV-2

Approach to CALR-Positive Myeloproliferative Neoplasms
CALR mutations are a revolutionary discovery and represent an important hallmark of myeloproliferative neoplasms (MPN), especially essential thrombocythemia and primary myelofibrosis. To date, several CALR mutations were identified, with only frameshift mutations linked to the diseased phenotype. It is of diagnostic and prognostic importance to properly define the type of CALR mutation and subclassify it according to its structural similarities to the classical mutations, a 52-bp deletion (type 1 mutation) and a 5-bp insertion (type 2 mutation), using a statistical approximation algorithm (AGADIR). Today, the knowledge on the pathogenesis of CALR-positive MPN is expanding and several cellular mechanisms have been recognized that finally cause a clonal hematopoietic expansion.

Prostate Cancer
Metastasis of prostate cancer often results in death of the patient. A cluster of fatty acid-binding protein (FABP) genes involved in transportation, accumulation and utilization of fatty acids are co-amplified and preferentially expressed in metastatic prostate cancer compared to localized disease. These genes, namely FABP12, FABP4, FABP9, FABP8 and FABP5, individually and collectively, promote properties associated with prostate cancer metastasis. Levels of these FABP genes may serve as an indicator of prostate cancer aggressiveness, and that inhibiting the action of FABP genes may provide a new approach to prevent and/or treat metastatic prostate cancer.

GABAAR-Mediated Currents
Propofol or barbiturate application at low concentrations increases desensitization and slows deactivation of GABA-induced current and propofol/barbiturate at high concentrations directly elicits after-responses upon their washout in hippocampal or sensory neurons. It is postulated that the generation of such after-responses is caused by removal of the blockade by anesthetic agents as partial antagonists. However, the increased desensitization was invariably followed by slowdown of deactivation of GABA-induced current, and the after-response may arise as a consequence of extreme slowdown of deactivation following strong desensitization. It is thus possible that propofol and barbiturate can facilitate resensitization of GABA responses. Propofol and barbiturate are useful to treat the alcohol/benzodiazepine withdrawal syndrome. Considering that the slowdown of deactivation following desensitization and the after-response induced by propofol or barbiturate application, the regulatory mechanisms of desensitization/resensitization of GABAAR-mediated currents might be important for understanding the treatment of the alcohol/benzodiazepine withdrawal syndrome.
