Air China Flight 112
Air China Flight 112 was a scheduled international passenger flight on 15 March 2003 that carried a 72-year-old man infected with severe acute respiratory syndrome (SARS). This man would later become the index passenger for the infection of another 20 passengers and two aircraft crew, resulting in the dissemination of SARS north to inner Mongolia and south to Thailand. The incident demonstrated how a single person could spread disease via air travel and was one of a number of superspreading events in the global spread of SARS in 2003. The speed of air travel and the multidirectional routes taken by affected passengers accelerated the spread of SARS with a consequential response from the World Health Organization (WHO), the aviation industry and the public. The incident was atypical in that passengers sitting at a distance from the index passenger were affected and the flight was only three hours long. Until this event, it was thought that there was only a significant risk of infection in flights of more than eight hours duration and in just the two adjacent seating rows. Other flights at the time with confirmed passengers with SARS did not have the same extent of infection spread. Some experts have questioned the interpretation of the incident and highlighted that some passengers may have been infected already. The role of cabin air has also come under question and the incident involving Flight 112 has led to some experts calling for further research into patterns of airborne transmission on commercial flights.

Airway Inflammation in Asthma Pathogenesis
Asthma is a heterogeneous disease characterized by chronic airway inflammation. Group 2 innate lymphoid cells (ILC2) play an important role in the pathogenesis of asthma. ILC2s lack antigen-specific receptors and respond to epithelial-derived cytokines, leading to the induction of airway eosinophilic inflammation in an antigen-independent manner. Additionally, ILC2s might be involved in the mechanism of steroid resistance. Numerous studies in both mice and humans have shown that ILC2s induce airway inflammation through inflammatory signals, including cytokines and other mediators derived from immune or non-immune cells.

Airway Microbiota for Lung Cancer
Recent research on cancer-associated microbial communities has elucidated the interplay between bacteria, immune cells, and tumor cells; the bacterial pathways involved in the induction of carcinogenesis; and their clinical significance. Although accumulating evidence shows that a dysbiotic condition is associated with lung carcinogenesis, the underlying mechanisms remain unclear. Microorganisms possibly trigger tumor initiation and progression, presumably via the production of bacterial toxins and other pro-inflammatory factors. The purpose of this review is to discuss the basic role of the airway microbiome in carcinogenesis and the underlying molecular mechanisms, with the aim of developing anticancer strategies involving the airway microbiota. In addition, the mechanisms via which the microbiome acts as a modulator of immunotherapies in lung cancer are summarized.

Airways Type-2 Related Disorders
Chronic rhinosinusitis (CRS) has recently undergone a significant paradigm shift, moving from a phenotypical classification towards an “endotype-based” definition that places more emphasis on clinical and therapeutic aspects. Similar to other airway diseases, like asthma, most cases of CRS in developed countries exhibit a dysregulated type-2 immune response and related cytokines. Consequently, the traditional distinction between upper and lower airways has been replaced by a “united airway” perspective. Additionally, type-2 related disorders extend beyond respiratory boundaries, encompassing conditions beyond the airways, such as atopic dermatitis. This necessitates a multidisciplinary approach. Moreover, consideration of possible systemic implications is crucial, particularly in relation to sleep-related breathing diseases like Obstructive Sleep Apnoea Syndrome (OSAS) and the alteration of systemic inflammatory mediators such as nitric oxide. The trends in epidemiological, economic, and social burden are progressively increasing worldwide, indicating syndemic characteristics.

AK-I-190
Castration-resistant prostate cancer (CRPC) is a clinical challenge in treatment because of its aggressive nature and resistance to androgen deprivation therapy. Topoisomerase II catalytic inhibitors have been suggested as a strategy to overcome these issues. We previously reported AK-I-190 as a novel topoisomerase II inhibitor. In this study, the mechanism of AK-I-190 was clarified using various types of spectroscopic and biological evaluations. AK-I-190 showed potent topoisomerase II inhibitory activity through intercalating into DNA without stabilizing the DNA-enzyme cleavage complex, resulting in significantly less DNA toxicity than etoposide, a clinically used topoisomerase II poison. AK-I-190 induced G1 arrest and effectively inhibited cell proliferation and colony formation in combination with paclitaxel in an androgen receptor–negative CRPC cell line.
