This Entry Collection focuses on current progress in understanding the role of chromatin structure, its modifications and remodeling in developmental and physiological processes. Eukaryotic genomes are packed into the supramolecular nucleoprotein structure of chromatin. Therefore, our understanding of processes such as DNA replication and repair, transcription, and cell differentiation requires understanding the structure and function of chromatin. While the nucleotide sequence of the DNA component of chromatin constitutes the genetic material of the cell, the other chromatin components (and also modifications of bases in the DNA itself) participate in so-called epigenetic processes. These processes are essential, e.g., in ontogenesis or adaptation to environmental changes. Therefore, epigenetics is particularly important (and elaborated) in plants that show a high developmental plasticity and, as sessile organisms, display an enormous capacity to cope with environmental stress. In these processes, epigenetic mechanisms show a crosstalk with plant signaling pathways mediated by phytohormones and redox components. Sustainable Concrete Quality Management
The development of a concrete mixture design process for high-quality concrete production with sustainable values is a complex process because of the multiple required properties at the green/hardened state of concrete and the interdependency of concrete mixture parameters. A new multi-criteria decision making (MCDM) technique based on Technique of Order Preference Similarity to the Ideal Solution (TOPSIS) methodology is applied to a fuzzy setting for the selection of concrete mix factors and concrete mixture design methods with the aim towards sustainable concrete quality management. Three objective properties for sustainable quality concrete are adopted as criteria in the proposed MCDM model. The seven most dominant concrete mixture parameters with consideration to sustainable concrete quality issues, i.e., environmental (density, durability) and socioeconomic criteria (cost, optimum mixture ingredients ratios), are proposed as sub-criteria. Three mixture design techniques that have potentiality to include sustainable aspects in their design procedure, two advanced and one conventional concrete mixture design method, are taken as alternatives in the MCDM model. The proposed selection support framework may be utilized in updating concrete design methods for sustainability and in deciding the most dominant concrete mix factors that can provide sustainable quality management in concrete production as well as in concrete construction. The concrete mix factors found to be most influential to produce sustainable concrete quality include the water/cement ratio and density. The outcomes of the proposed MCDM model of fuzzy TOPSIS are consistent with the published literature and theory. TheDOE method was found to be more suitable in sustainable concrete quality management considering its applicable objective quality properties and concrete mix factors.

Multifaceted Mechanisms of Metformin
While there are various kinds of drugs for type 2 diabetes mellitus at present, in this review article, we focus on metformin which is an insulin sensitizer and is often used as a first-choice drug worldwide. Metformin mainly activates adenosine monophosphate-activated protein kinase (AMPK) in the liver which leads to suppression of fatty acid synthesis and gluconeogenesis. Metformin activates AMPK in skeletal muscle as well, which increases translocation of glucose transporter 4 to the cell membrane and thereby increases glucose uptake. Further, metformin suppresses glucagon signaling in the liver by suppressing adenylate cyclase which leads to suppression of gluconeogenesis. In addition, metformin reduces autophagy failure observed in pancreatic β-cells under diabetic conditions. Furthermore, it is known that metformin alters the gut microbiome and facilitates the transport of glucose from the circulation into excrement. It is also known that metformin reduces food intake and lowers body weight by increasing circulating levels of the peptide hormone growth/differentiation factor 15 (GDF15). Furthermore, much attention has been drawn to the fact that the frequency of various cancers is lower in subjects taking metformin. Metformin suppresses the mechanistic target of rapamycin (mTOR) by activating AMPK in pre-neoplastic cells, which leads to suppression of cell growth and an increase in apoptosis in pre-neoplastic cells. It has been shown recently that metformin consumption potentially influences the mortality in patients with type 2 diabetes mellitus and coronavirus infectious disease (COVID-19). Taken together, metformin is an old drug, but multifaceted mechanisms of action of metformin have been unraveled one after another in its long history.

Diatom frustules are considered as a sustainable source for several industrial applications because of their high biocompatibility and the easiness of surface functionalisation, which make frustules suitable for regenerative medicine and as drug carriers. Frustules are made of hydrated silica, and can be extracted and purified both from living and fossil diatoms using acid treatments or high temperatures. Biosilica frustules have proved to be suitable for biomedical applications, but, unfortunately, they are not officially recognised as safe by governmental food and medical agencies yet.

Unlocking loxP to Track Genome
The development of CRISPR-associated proteins, such as Cas9, has led to increased accessibility and ease of use in genome editing. However, additional tools are needed to quantify and identify successful genome editing events in living animals. We developed a method to rapidly quantify and monitor gene editing activity non-invasively in living animals that also facilitates confocal microscopy and nucleotide level analyses. Here we report a new CRISPR “fingerprinting” approach to activating luciferase and fluorescent proteins in mice as a function of gene editing. This system is based on experience with our prior cre recombinase (cre)-detector system and is designed for Cas editors able to target loxP including gRNAs for SaCas9 and ErCas12a. These CRISPRs cut specifically within loxP, an approach that is a departure from previous gene editing in vivo activity detection techniques that targeted adjacent stop sequences. In this sensor paradigm, CRISPR activity was monitored non-invasively in living cre reporter mice (FVB.129S6(B6)-Gt(ROSA)26Sortm1(Luc)Kael/J and Gt(ROSA)26Sortm4(ACTB-tdTomato,-EGFP)Luo/J, which will be referred to as LSL-luciferase and mT/mG throughout the paper) after intramuscular or intravenous hydrodynamic plasmid injections, demonstrating utility in two diverse organ systems. The same genome-editing event was examined at the cellular level in specific tissues by confocal microscopy to determine the identity and frequency of successfully genome-edited cells. Further, SaCas9 induced targeted editing at efficiencies that were comparable to cre, demonstrating high effective delivery and activity in a whole animal.

The use of biological templates for the suitable growth of adipose-derived mesenchymal stem cells (AD-MSC) and “neo-tissue” construction has exponentially increased over the last years. The bioengineered scaffolds still have a prominent and biocompatible framework playing a role in tissue regeneration. In order to supply AD-MSCs, biomaterials, as the stem cell niche, are more often supplemented by or stimulate molecular signals that allow differentiation events into several strains, besides their secretion of cytokines and effects of immunomodulation. This systematic review aims to highlight the details of the integration of several types of biomaterials used in association with AD-MSCs, collecting notorious and basic data of in vitro and in vivo assays, taking into account the relevance of the interference of the cell lineage origin and handling cell line protocols for both the replacement and repairing of damaged tissues or organs in clinical application. Our group analyzed the quality and results of the 98 articles selected from PubMed, Scopus and Web of Science. A total of 97% of the articles retrieved demonstrated the potential in clinical applications. The synthetic polymers were the most used biomaterials associated with AD-MSCs and almost half of the selected articles were applied on bone regeneration.
