Immune Checkpoint Therapy Resistance with SHP2 Inhibition
SHP2 (Src Homology 2 Domain-Containing Phosphatase 2) is a protein tyrosine phosphatase widely expressed in various cell types. SHP2 plays a crucial role in different cellular processes, such as cell proliferation, differentiation, and survival. Aberrant activation of SHP2 has been implicated in multiple human cancers and is considered a promising therapeutic target for treating these malignancies. The PTPN11 gene and functions encode SHP2 as a critical signal transduction regulator that interacts with key signaling molecules in both the RAS/ERK and PD-1/PD-L1 pathways; SHP2 is also implicated in T-cell signaling. SHP2 may be inhibited by molecules that cause allosteric (bind to sites other than the active site and attenuate activation) or orthosteric (bind to the active site and stop activation) inhibition or via potent SHP2 degraders. These inhibitors have anti-proliferative effects in cancer cells and suppress tumor growth in preclinical models. In addition, several SHP2 inhibitors are in clinical trials for cancer treatment.

Oxaliplatin-Induced Peripheral Neuropathy
Oxaliplatin-induced peripheral neurotoxicity (OIPN) is a severe and potentially permanent side effect of cancer treatment affecting the majority of oxaliplatin-treated patients, mostly with the onset of acute symptoms, but also with the establishment of a chronic sensory loss that is supposed to be due to dorsal root ganglia neuron damage. The pathogenesis of OIPN is still largely unknown. This lack of information is a limitation in the identification of effective strategies to prevent or limit OIPN incidence and severity. So far, no treatment is available for the prevention of OIPN, although duloxetine showed moderate evidence of efficacy in the reduction of OIPN symptoms. On this background, several neuroprotection clinical trials are ongoing in oxaliplatin-treated patients, although only part of them relies on solid preclinical evidence supporting the study hypothesis. Based on the available literature it can be concluded that dose and schedule modification is currently the most effective approach to limit the severity of OIPN since pharmacological prevention and treatment of OIPN still remains an unmet clinical need.

Water–Energy Nexus
The sustainability or green building rating system used in the United Arab Emirates (UAE), Pearl Rating System (PRS) of Estidama, similar to other international rating systems such as LEED and BREEAM, considers several strategies, regulations, and policies to improve the energy and water performance in buildings. However, the applicability of considering water as part of energy or the fact that utilization of energy mandates the usage of water seems unexplored and not yet included in any of the existing building rating systems including LEED. A unified approach of the water and energy resources is hence vital for future considerations in energy policy, planning and the inclusion of the same in the sustainability rating systems as well. This paper investigates, as a case study, the prospects of water-energy nexus in the prevailing UAE green building rating system; Pearl Rating System(PRS) to bring out if any water conservation strategy has an adverse effect on energy and vice versa. The review revealed that the major shortcomings of PRS in terms of water-energy nexus strategy are the usage of reference codes not suitable to UAE’s climate and geographical conditions, inexistent synergy between some credit categories, oversight of rebound effects, and a need for credit reassessment. The paper also recommends that any proposed strategy to realign credit categories in terms of water-energy nexus with the potential risk to also have a hidden negative rebound effect which the researchers and practitioners should identify lest the water- energy tradeoff bring unprecedented repercussions. The theoretical analysis established that the bifurcating management of water and energy in the sustainability rating system and energy policy needs to be revisited in order to reap more sustainable and optimum results that is environmentally, ecologically and financially consistent. Vitamin D, microbiome, and IBD
Inflammatory bowel disease (IBD) is a chronic inflammation of the gastrointestinal tract (GIT), including Crohn’s disease (CD) and ulcerative colitis (UC), which differ in the location and lesion extensions. Both diseases are associated with microbiota dysbiosis, with a reduced population of butyrate-producing species, abnormal inflammatory response, and micronutrient deficiency (e.g., vitamin D hypovitaminosis). Vitamin D (VitD) is involved in immune cell differentiation, gut microbiota modulation, gene transcription, and barrier integrity. Vitamin D receptor (VDR) regulates the biological actions of the active VitD (1α,25-dihydroxyvitamin D3), and is involved in the genetic, environmental, immune, and microbial aspects of IBD. VitD deficiency is correlated with disease activity and its administration targeting a concentration of 30 ng/mL may have the potential to reduce disease activity. Moreover, VDR regulates functions of T cells and Paneth cells and modulates release of antimicrobial peptides in gut microbiota-host interactions. Meanwhile, beneficial microbial metabolites, e.g., butyrate, upregulate the VDR signaling.

Energy Consumption of Vacuum Freeze Dryer
The vacuum freeze-drying process is characterized by sublimating the frozen object into a solid state in a vacuum environment, sublimating the water from the object, and finally drying it. At the same time, sublimating the water into the cold trap coil to re form ice. This characteristic determines that the freeze-drying machine needs to use the heat medium circulation system to heat the material to sublimate the water of the object, The ice condensed on the cold trap coil also needs a lot of heat to melt and discharge. During the freeze-drying process of the freeze dryer, the cold trap needs to continuously capture the water vapor generated by sublimation to freeze the cold trap coil. The ice captured by the cold trap coil needs to be melted after each batch of freeze-drying, which in turn requires a large amount of high-temperature defrosting water or thawing steam. For the freeze dryer with heat medium circulation system, sublimation drying also needs to continuously absorb heat, The heat required for the whole process is provided by the electric heater of the heat medium circulation system. The preparation of defrosting water or defrosting steam and the electric heater that provides necessary heat for sublimation require a lot of energy consumption. The freeze dryer consumes too much energy, the freeze-drying cost is high and the energy waste is serious.

Nutritional Therapies on the Intestinal Microbiome
Inflammatory bowel disease (IBD) is a chronic, autoimmune disorder of the gastrointestinal tract with numerous genetic and environmental risk factors. Patients with Crohn’s disease (CD) or ulcerative colitis (UC) often demonstrate marked disruptions of their gut microbiome. The intestinal microbiota is strongly influenced by diet. The association between the increasing incidence of IBD worldwide and increased consumption of a westernized diet suggests host nutrition may influence the progression or treatment of IBD via the microbiome. Several nutritional therapies have been studied for the treatment of CD and UC. While their mechanisms of action are only partially understood, existing studies do suggest that diet-driven changes in microbial composition and function underlie the diverse mechanisms of nutritional therapy. Despite existing therapies for IBD focusing heavily on immune suppression, nutrition is an important treatment option due to its superior safety profile, potentially low cost, and benefits for growth and development. These benefits are increasingly important to patients. In this entry, we will describe the clinical efficacy of the different nutritional therapies that have been described for the treatment of CD and UC.

Psychological Symptoms in COVID-19 Patients
There is growing evidence of studies associating COVID-19 survivors with increased mental health consequences. Mental health implications related to a COVID-19 infection include both acute and long-term consequences. Here we discuss COVID-19-associated psychiatric sequelae, particularly anxiety, depression, and post-traumatic stress disorder (PTSD), drawing parallels to past coronavirus outbreaks. A literature search was completed across three databases, using keywords to search for relevant articles. The cause may directly correlate to the infection through both direct and indirect mechanisms, but the underlying etiology appears more complex and multifactorial, involving environmental, psychological, and biological factors. Although most risk factors and prevalence rates vary across various studies, being of the female gender and having a history of psychiatric disorders seem consistent. Several studies will be presented, demonstrating COVID-19 survivors presenting higher rates of mental health consequences than the general population. The possible mechanisms by which the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) enters the brain, affecting the central nervous system (CNS) and causing these psychiatric sequelae, will be discussed, particularly concerning the SARS-CoV-2 entry via the angiotensin-converting enzyme 2 (ACE-2) receptors and the implications of the immune inflammatory signaling on neuropsychiatric disorders. Some possible therapeutic options will also be considered.
