Alpha-Amylase Inhibitory Metabolites from Pericarps of Garcinia mangostana
Two new benzophenones and five known metabolites were purified from Garcinia mangostana ((GM) pericarps. They were characterized by various spectral techniques. Compounds 6 and 7 displayed AAI activity. They also exhibited highly negative docking scores, when complexed with 5TD4. Their complexes with the α-amylase were found to be stable over the course of 50 ns. These results supported the previous reports that GM can potentially represent an appealing treatment of diabetes and its related disorders.

Glucosamine and Chondroitin for Osteoarthritis
Glucosamine is a water-soluble amino monosaccharide available in two forms (glucosamine sulphate and glucosamine hydrochloride), which is a normal constituent of glycosaminoglycans (GAGs) in cartilage matrix and in the synovial fluid and consequently present in high quantities in articular cartilage. It is a constituent of keratan sulphate. Chondroitin is a major component of the extracellular matrix of articular cartilage, which played an important role in creating considerable osmotic pressure. In this way, it could provide cartilage with resistance and elasticity to resist tensile stresses during loading conditions.

Metformin in Diabetic and Non-Diabetic Bone Impairment
Metformin is a widely-used anti-diabetic drug in patients with type 2 diabetic mellitus (T2DM) due to its safety and efficacy in clinical. The classic effect of metformin on lowering blood glucose levels is to inhibit liver gluconeogenesis that reduces glucose production as well as increases peripheral glucose utilization. However, the factors such as hyperglycemia, insulin deficiency, reduced serum levels of insulin-like growth factor-1 (IGF-1) and osteocalcin, accumulation of advanced glycation end products (AGEs), especially in collagen, microangiopathy, and inflammation reduced bone quality in diabetic patients. However, hyperglycemia, insulin deficiency, reduced levels of insulin-like growth factor-1 (IGF-1) and osteocalcin in serum, accumulation of advanced glycation end products (AGEs) in collagen, microangiopathy, and inflammation, reduce bone quality in diabetic patients. Furthermore, the imbalance of AGE/RAGE results in bone fragility via attenuating osteogenesis. Thus, adequate glycemic control by medical intervention is necessary to prevent bone tissue alterations in diabetic patients. Metformin mainly activates adenosine 5′ -monophosphate-activated protein kinase (AMPK), and inhibits mitochondrial respiratory chain complex I in bone metabolism. In addition, metformin increases the expression of transcription factor runt-related transcription factor2 (RUNX2) and Sirtuin protein to regulate related gene expression in bone formation. Until now, there are a lot of preclinical or clinical findings on the application of metformin to promote bone repair. Taken together, metformin is considered as a potential medication for adjuvant therapy in bone metabolic disorders further to its antidiabetic effect. Taken together, as a conventional hypoglycemia drug with multifaceted effects, metformin has been considered a potential adjuvant drug for the treatment of bone metabolic disorders.

Highly Selective Antagonist of GluK3 Kainate Receptor Subtype
Kainate receptors belong to the family of glutamate receptors ion channels, which are responsible for the majority of rapid excitatory synaptic transmission in the central nervous system. The therapeutic potential of kainate receptors is still poorly understood, which is also due to the lack of potent and subunit-selective pharmacological tools. In search of selective ligands for the GluK3 kainate receptor subtype, a series of quinoxaline-2,3-dione analogues was synthesized and pharmacologically characterized at selected recombinant ionotropic glutamate receptors. Among them, compound 28 was found to be a competitive GluK3 antagonist with submicromolar affinity and unprecedented high binding selectivity, showing a 400-fold preference for GluK3 over other homomeric receptors GluK1, GluK2, GluK5 and GluA2.

Delivering drugs to horses orally is a major challenge, as the physiology and anatomy of the equine gastro-intestinal tract (GIT) can result in poor bioavailability of some oral drugs. Additionally, due to their size, the dosages required are much larger and higher cost compared to those of humans; with potential of adverse effects, especially for antibiotics, following the administration of some drugs.
