This  is a comparative study that explores  Indigenous communities, rural poverty, and mental health and well-being in Australia and Canada. It draws upon a person-centred approach  as described by Carl Rogers' humanistic psychology framework. Hence, the role of women within these communities and their strength  in support of the well-being of their families is also examined. Themes that emerge within a humanistic theoretical framework will be highlighted. Examination of current data is provided and existing research findings are compared where relevant to indigenous populations.

Bacterial pathogens are serious causative agents of infectious disease. Such microorganisms are resistant to multiple antimicrobial agents, thereby compromising the therapeutic efficacy of treatment.  Multidrug-resistant pathogens harbor antimicrobial efflux pumps, many transporters of which are members of the extensive major facilitator superfamily of proteins. These bacterial multidrug efflux pumps are good molecular targets for modulation and possible inhibition.  This entry briefly discusses several current developments for drug efflux pump modulation.

Infectious Diseases Associated with and Causing Disaster
In 2019, 396 natural disasters were recorded in the Emergency Events Database (EM-DAT), with 11,755 deaths, 95 million people affected, and USD 103 billion in economic losses worldwide. This burden was not shared equally since Asia suffered the highest impact, accounting for 40% of disaster events, 45% of deaths and 74% of the total affected. During disasters, a lack of safe water access and inadequate sanitation facilities allow the transmission of water-borne and food-borne pathogens. Diarrheal diseases such as cholera, typhoid fever, and shigellosis cause epidemics with high mortality rates. Malaria and other vector-borne diseases in risk areas include arboviruses, such as dengue, yellow fever, Japanese encephalitis, Rift Valley fever, and tick-borne illnesses, including Crimean–Congo hemorrhagic fever and typhus. Diseases associated with overcrowding, such as measles in unvaccinated areas and tuberculosis, can occur after natural disasters.

Reinventing approved therapeutic proteins for a new dose, a new formulation, a new route of administration, an improved safety profile, a new indication, or a new conjugate with a drug or a radioactive source is a creative approach to benefit from the billions spent on developing new therapeutic proteins. These new opportunities were created only recently with the arrival of Artificial Intelligence (AI)/Machine Learning (ML) tools and high throughput screening technologies. Furthermore, the complex nature of proteins offers mining opportunities that are not possible with chemical drugs; bringing in newer therapies without spending billions makes this path highly lucrative financially while serving the dire needs of humanity.

Retinal Organization
The retinal arhitecture is similar across all vertebrates despite the morphological and functional peculiarities. The retina is composed of two main parts: the single-layred retinal pigment epithelium and the multilayred neuroretina which includes a number of neuronal and glial cell types. Homeobox genes from different classes are accepted as critical for eye field specification and retinal cells type differentiation by a broad array of loss- or gain-of-function models. Among these genes are some that are known to cause inherited retinal diseases (IRDs) that disturb the formation, function, and survival of rod and cone photoreceptors, ganglion cells, or retinal pigment epithelial cells [4,6–8]. The advances in the field of genetics and high-throughput next-generation sequencing and cell technologies allow for deepening of knowledge of the genetic basis of inherited retinal diseases (IRDs), as well as improve their diagnostics and therapy.

Approach to CALR-Positive Myeloproliferative Neoplasms
CALR mutations are a revolutionary discovery and represent an important hallmark of myeloproliferative neoplasms (MPN), especially essential thrombocythemia and primary myelofibrosis. To date, several CALR mutations were identified, with only frameshift mutations linked to the diseased phenotype. It is of diagnostic and prognostic importance to properly define the type of CALR mutation and subclassify it according to its structural similarities to the classical mutations, a 52-bp deletion (type 1 mutation) and a 5-bp insertion (type 2 mutation), using a statistical approximation algorithm (AGADIR). Today, the knowledge on the pathogenesis of CALR-positive MPN is expanding and several cellular mechanisms have been recognized that finally cause a clonal hematopoietic expansion.

NK Cells in Cancer Therapy
       Cellular therapy has emerged as an attractive option for the treatment of cancer, and adoptive transfer of chimeric antigen receptor (CAR) expressing T cells has gained FDA approval in hematologic malignancy. However, limited efficacy was observed using CAR-T therapy in solid tumors. Natural killer (NK) cells are crucial for tumor surveillance and exhibit potent killing capacity of aberrant cells in an antigen-independent manner. Adoptive transfer of unmodified allogeneic or autologous NK cells has shown limited clinical benefit due to factors including low cell number, low cytotoxicity and failure to migrate to tumor sites. To address these problems, immortalized and autologous NK cells have been genetically engineered to express high affinity receptors (CD16), CARs directed against surface proteins (PD-L1, CD19, Her2, etc.) and endogenous cytokines (IL-2 and IL-15) that are crucial for NK cell survival and cytotoxicity, with positive outcomes reported by several groups both preclinically and clinically. With a multitude of NK cell-based therapies currently in clinic trials, it is likely they will play a crucial role in next-generation cell therapy-based treatment. In this review, we will highlight the recent advances and limitations of allogeneic, autologous and genetically enhanced NK cells used in adoptive cell therapy.

Fc-Engineered Antibodies
Monoclonal antibody (mAb) therapy has rapidly changed the field of cancer therapy. In 1997, the CD20-targeting mAb rituximab was the first mAb to be approved by the U.S. Food and Drug Administration (FDA) for treatment of cancer. Within two decades, dozens of mAbs entered the clinic for treatment of several hematological cancers and solid tumors, and numerous more are under clinical investigation. The success of mAbs as cancer therapeutics lies in their ability to induce various cytotoxic machineries against specific targets. These cytotoxic machineries include antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and complement-dependent cytotoxicity (CDC), which are all mediated via the fragment crystallizable (Fc) domain of mAbs. In this review article we will outline the novel approaches of engineering these Fc domains of mAbs to enhance their Fc-effector function and thereby their anti-tumor potency, with specific focus to summarize their (pre-) clinical status for the treatment of B-cell malignancies, including chronic lymphocytic leukemia (CLL), B-cell Non-Hodgkin lymphoma (B-NHL), and multiple myeloma (MM).
