Polyphenolic-Loaded Nanocarriers for Inflammation-Mediated Diseases
The global increase and prevalence of inflammatory-mediated diseases have been a great menace to human welfare. Several works have demonstrated the anti-inflammatory potentials of natural polyphenolic compounds, including flavonoid derivatives (EGCG, rutin, apigenin, naringenin) and phenolic acids (GA, CA, etc.), among others (resveratrol, curcumin, etc.). In order to improve the stability and bioavailability of these natural polyphenolic compounds, their recent loading applications in both organic (liposomes, micelles, dendrimers, etc.) and inorganic (mesoporous silica, heavy metals, etc.) nanocarrier technologies are being employed.

The altered functioning of the Wnt signaling in human tumors points to the therapeutic strategy of the inhibition of its activity. Drugging the Wnt signaling pathway continues to be one of the promising approaches for future tumor treatment, both alone and in combination therapy that would impact the clinical outcomes and survival of patients.

Type 2 diabetes mellitus (T2DM) is the metabolic disease with the highest morbidity rates worldwide. The condition is characterized by hyperglycemia, insulin resistance, hyperlipidemia, and chronic inflammation, among other detrimental conditions. These decrease the efficiency of the immune system, leading to an increase in the susceptibility to bacterial infections. Maintaining an optimal blood glucose level is crucial in relation to the treatment of T2DM, because if the level of this carbohydrate is lowered, the risk of infections can be reduced.

Tregs in Psoriasis
Psoriasis is a chronic inflammatory skin disease, which affects 2–4% of the population worldwide. Psoriasis has a strong genetic component, but environmental factors are also important. T-cell derived factors such as Psoriasis is characteristic, with thickening and scaling of the epidermisdue to hyper-proliferation of keratinocytes (acanthosis). CD4 and CD8 T-cell infiltrate characterizes the skin in psoriasis, a T-cell-driven disease, but other cell types are also present in the lesions, including neutrophils, macrophages, NK cells, dendritic cells. It has been long known that the pathogenesis of psoriasis is driven by T-cell derived factors, produced by subsets including T-helper (Th) 1 cells, Th17, Th22 and regulatory T cells (Tregs).

Efficacy of Elexacaftor-Tezacaftor-Ivacaftor in Cystic Fibrosis Treatment
Elexacaftor/Tezacaftor/Ivacaftor (ELX/TEZ/IVA) is a new CFTR (Cystic Fibrosis Transmembrane Conductance Regulator) modulator treatment, which has shown an improvement in different clinical outcomes in patients with cystic fibrosis (CF). ELX/TEZ/IVA has a significant positive effect on the lung function of patients with CF, by ameliorating parameters such as FEV1, LCI, pulmonary exacerbations or sweat chloride concentration, increasing BMI and improving quality of their life. Its role in cystic fibrosis-related diabetes (CFRD) is not yet clear. It was found that this new CFTR modulator has an overall favorable safety profile, with mild to moderate adverse events.

PARK7/DJ-1 in Gut-Brain Axis Diseases
Parkinson’s disease 7 (PARK7/DJ-1) is a multifunctional protein whose protective role has been widely demonstrated in neurodegenerative diseases, including PD, AD, or ischemic stroke. Recent studies also revealed the importance of PARK7/DJ-1 in the maintenance of the gut microbiome and also in the regulation of intestinal inflammation. All these findings suggest that PARK7/DJ-1 may be a link and also a potential therapeutic target in gut and brain diseases.
