Synthetic Cathinones and Synthetic Cannabinoids
Novel psychoactive substances (NPS) are compounds of natural and synthetic origin, similar to traditional drugs of abuse. NPS are involved in a contemporary trend whose origin lies in a thinner balance between legitimate therapeutic drug research and legislative control. The contemporary NPS trend resulted from the replacement of MDMA by synthetic cathinones in ‘ecstasy’ during the 2000s. The most common NPS are synthetic cannabinoids and synthetic cathinones. Interestingly, during the last 50 years, these two classes of NPS have been the object of scientific research for a set of health conditions.

Synthesis of the Coumarin Core
Coumarin compounds are attractive organic compounds with many practical applications. Among them there are compounds with biological activity, pharmaceuticals, agrochemicals, dyes, and optoelectronic materials. For this reason, enormous and continuous attempts were made to develop new synthetic pathways and protocols to facilitate the key cyclization reaction of heterocyclic ring and its regioselective functionalization. Of the numerous proposed reactions for the preparation of coumarins, those based on transition metal catalysts have been frequently used recently. Such processes as intramolecular and intermolecular hydroarylation of alkenes or alkynes can be mentioned among the most effective reactions proceeding via the activation of the C–H bond. Knowledge about the mechanistic foundations of catalytic processes seems to be of significant importance in order to improve them and simplify the conditions. Direct functionalization of the coumarin skeleton seems to be one of the more difficult tasks in recent times.

N-heterocycles, both saturated and unsaturated, are ubiquitous biologically active molecules that are extremely appealing scaffolds in drug discovery programs. Although classical synthetic methods have been developed to access many relevant N-heterocyclic scaffolds, representing well-established and reliable routes, some do not meet the needs of sustainability. In this context, several advances have been made towards the sustainable synthesis of N-heterocycles.

Symmetrical Compounds in Insulin-Sensitizing Treatments for Type 2 Diabetes
Type 2 diabetes is a complex disease that involves damage to multiple signaling pathways. Through various pathways, chronic high blood glucose generates or aggravates insulin resistance. The drugs most widely used to decrease insulin resistance without producing hypoglycemia are biguanides (e.g., metformin) and thiazolidinediones (TZDs, such as pioglitazone). Symmetrical molecules have been a focus of research due to their unique structural characteristics, including stability and internal balance. Some advantages and disadvantages of utilizing symmetrical and asymmetrical thiazolidinediones as insulin sensitizers (or even antioxidant molecules) are mentioned.

Sustained Antiviral Ocular Drug Delivery
The eyes are the window to the world and the key to communication, but they are vulnerable to multitudes of ailments. More serious than is thought, corneal infection by herpes simplex viruses (HSVs) is a prevalent yet silent cause of blindness in both the paediatric and adult population, especially if immunodeficient. Globally, there are 1.5 million new cases and forty thousand visual impairment cases reported yearly. The Herpetic Eye Disease Study recommends topical antiviral as the front-line therapy for HSV keratitis. Ironically, topical eye solutions undergo rapid nasolacrimal clearance, which necessitates oral drugs but there is a catch of systemic toxicity. The hurdle of antiviral penetration to reach an effective concentration is further complicated by drugs’ poor permeability and complex layers of ocular barriers.

Superficial Heat Therapy
Low back pain (LBP) is an exceptionally common musculoskeletal problem and a leading cause of disability. LBP is experienced by most adults at some stage of their life, with an estimated 577 million people affected in 2017. A recent systematic literature review stated that the prevalence of LBP ranges from 1.4% to 20.0%, and the incidence from 0.024% to 7.0%. The review focused on the application and effect of mild superficial heat for musculoskeletal pain, especially back pain. The review was able to show that the application of mild heat is easily applicable and effective when indicated.

Sulforaphane (SFN) is a natural glucosinolate found in cruciferous vegetables that acts as a chemopreventive agent, but its mechanism of action is not clear. Due to antioxidative mechanisms being thought central in preventing cancer progression, SFN could play a role in oxidative processes. Since redox imbalance with increased levels of reactive oxygen species (ROS) is involved in the initiation and progression of bladder cancer, this mechanism might be involved when chemoresistance occurs.
