Diet-Derived Phytochemicals Modulate the Gut Microbiome
Diet-derived phytochemicals modulate microbiome that is found to offer significant protective effects against colorectal cancer (CRC). The person’s lifestyle and the eating pattern have significant impacts on the CRC in a positive and/or negative way. Phytochemicals are a concoction of various bioactive compounds directing various cell signaling pathways that altered gut microbiota composition. This may support to destroy malignant cells with minor risks of emerging drug resistance. The effectiveness of CRC can be reduced by the use of various dietary phytochemicals or modulating microbiome that reduces or inverses the progression of a tumor, which could be promising and efficient in reducing the burden of CRC. Phytochemicals with modulation of gut microbiome continue to be auspicious investigations in CRC through noticeable anti-tumorigenic effects, which provides new openings for cancer inhibition and treatment.

Pulse Wave Velocity
Pulse wave velocity (PWV) is a simple, reproducible and non-invasive technique to assess arterial stiffness. It estimates the velocity of arterial wave propagation to travel a known distance between two anatomic sites within the arterial system [1]. PWV has been established as an independent predictor of cardiovascular mortality and is consequently used for diagnosis and prognosis in patients at risk of atherosclerotic cardiovascular disease [2]; however, PWV's significance relies on the fact that arterial stiffness, specially of muscular arteries can be modified by interventions that improve endothelial function such as exercise training, which makes PWV an important tool for cardiovascular risk management [3].

These alterations are mostly reversible with treatment discontinuation, except for priapism, which can require surgical interven-tion in some cases.

Bile Acids in the Liver and Gut Metabolism
Bile acids (BAs) represent heterogeneous amphipathic molecules that have both polar (water-soluble or hydrophilic) and apolar (water-insoluble or hydrophobic) parts, and therefore they can dissolve in water as well as in fat. The regulation of cholesterol homeostasis remains their main role in the host. The better water solubility in comparison with their precursor promotes cholesterol removal from the human body. The proper bile ratio of BAs and cholesterol prevents cholesterol precipitation and further formation of gallstones; therefore, the loss of BAs increases the risk of cholesterol stone development.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive solid malignancies. The complex mechanism underlying the development and progression of PDAC includes interactions between genomic, epigenomic, and signaling pathway alterations. PDAC remains a devastating disease, despite extensive research efforts focused on the discovery of early diagnostic biomarkers and efficient therapeutic approaches, showing a high resistance to treatment that significantly contributes to poor survival. The success of epi-drugs in hematological malignancies and the robust body of evidence supporting the hypothesis about their ability to synergize with other anti-cancer drugs, sensitizing resistant tumor cells in preclinical models, have encouraged the search for these agents suitable for the future clinical management of PDAC.

Anti-Inflammatory and Antiviral Effects of Cannabinoids
The COVID-19 pandemic caused by the SARS-CoV-2 virus made it necessary to search for new options for both causal treatment and mitigation of its symptoms. Scientists and researchers around the world are constantly looking for the best therapeutic options. These difficult circumstances have also spurred the re-examination of the potential of natural substances contained in Cannabis sativa L. Cannabinoids, apart from CB1 and CB2 receptors, may act multifacetedly through a number of other receptors, such as the GPR55, TRPV1, PPARs, 5-HT1A, adenosine and glycine receptors. The complex anti-inflammatory and antiviral effects of cannabinoids have been confirmed by interactions with various signaling pathways. Considering the fact that the SARS-CoV-2 virus causes excessive immune response and triggers an inflammatory cascade, and that cannabinoids have the ability to regulate these processes, it can be assumed that they have potential to be used in the treatment of COVID-19.

β-Caryophyllene and Rheumatoid Arthritis
β-caryophyllene (BCP) is a cannabinoid receptor 2 (CB2) agonist that tempers inflammation. An interaction between the CB2 receptor and peroxisome proliferator-activated receptor gamma (PPAR-γ) has been suggested and PPAR-γ activation exerts anti-arthritic effects. The aim of this study was to characterize the therapeutic activity of BCP and to investigate PPAR-γ involvement in a collagen antibody induced arthritis (CAIA) experimental model. CAIA was induced through intraperitoneal injection of a monoclonal antibody cocktail and lipopolysaccharide (LPS; 50 µg/100 µL/ip). CAIA animals were then randomized to orally receive either BCP (10 mg/kg/100 µL) or its vehicle (100 µL of corn oil). BCP significantly hampered the severity of the disease, reduced relevant pro-inflammatory cytokines, and increased the anti-inflammatory cytokine IL-13. BCP also decreased joint expression of matrix metalloproteinases 3 and 9. Arthritic joints showed increased COX2 and NF-kB mRNA expression and reduced expression of the PPARγ coactivator-1 alpha, PGC-1α, and PPAR-γ. These conditions were reverted following BCP treatment. Finally, BCP reduced NF-kB activation and increased PGC-1α and PPAR-γ expression in human articular chondrocytes stimulated with LPS. These effects were reverted by AM630, a CB2 receptor antagonist. These results suggest that BCP ameliorates arthritis through a cross-talk between CB2 and PPAR-γ.
