Herbal Approach, Bioactive Compounds, and Mechanism of Obesity
Obesity is arising as a global pandemic throughout the world. Obesity has tripled worldwide, creating an alarming situation. The majority of people nowadays are suffering from obesity and overweight. It affects health of people of all age groups, ethnicity, gender, and sex, and is linked to a sedentary lifestyle of people, poor eating habits, and disturbed sleeping patterns. It causes several diseases such as diabetes mellitus type 2, hypertension, cardiovascular diseases, asthma, gallstones, and colon cancer. Many synthetic anti-obesity drugs such as orlistat, lorcaserin, phentermine, bupropion, and liraglutide are already available on the market. However, these drugs have side effects, including dry mouth and sleeping disorders, dizziness, blood pressure, heart rate elevation, constipation, and headache. Humans have a long and ancient history of dependency on traditional medicinal plants and their major bioactive antioxidant components, such as quercetin, anthocyanins, and ellagic acid, for treating such diseases and disorders.

β-Hemoglobinopathies
β-hemoglobinopathies are the most common genetic disorders worldwide and are caused by mutations affecting the production or the structure of adult hemoglobin. Patients affected by these diseases suffer from anemia, impaired oxygen delivery to tissues, and multi-organ damage. In the absence of a compatible donor for allogeneic bone marrow transplantation, the lifelong therapeutic options are symptomatic care, red blood cell transfusions and pharmacological treatments. The last decades of research established lentiviral-mediated gene therapy as an efficacious therapeutic strategy. However, this approach is highly expensive and associated with a variable outcome depending on the effectiveness of the viral vector and the quality of the cell product. In the last years, genome editing emerged as a valuable tool for the development of curative strategies for β-hemoglobinopathies. Moreover, due to the wide range of its applications, genome editing has been extensively used to study regulatory mechanisms underlying globin gene regulation allowing the identification of novel genetic and pharmacological targets.

Intravesicular pH
Intravesicular pH plays a crucial role in melanosome maturation and function. Melanosomal pH changes during maturation from very acidic in the early stages to neutral in late stages. Neutral pH is critical for providing optimal conditions for the rate-limiting, pH-sensitive melanin-synthesizing enzyme tyrosinase (TYR). This dramatic change in pH is thought to result from the activity of several proteins that control melanosomal pH.

Gut Microbiota and Parkinson’s Disease
Parkinson’s disease (PD) is the second-most prevalent neurodegenerative or neuropsychiatric disease, affecting 1% of seniors worldwide. The gut microbiota (GM) is one of the key access controls for most diseases and disorders. Disturbance in the GM creates an imbalance in the function and circulation of metabolites, resulting in unhealthy conditions. Any dysbiosis could affect the function of the gut, consequently disturbing the equilibrium in the intestine, and provoking pro-inflammatory conditions in the gut lumen, which send signals to the central nervous system (CNS) through the vagus enteric nervous system, possibly disturbing the blood–brain barrier. The neuroinflammatory conditions in the brain cause accumulation of α-syn, and progressively develop PD. An important aspect of understanding and treating the disease is access to broad knowledge about the influence of dietary supplements on GM.

Adult Hippocampal Neurogenesis
Newborn neurons in the adult hippocampus are regulated by many intrinsic and extrinsic cues. It is well accepted that elevated glucocorticoid levels lead to downregulation of adult neurogenesis, which this review discusses as one reason why psychiatric diseases, such as major depression, develop after long-term stress exposure. In reverse, adult neurogenesis has been suggested to protect against stress-induced major depression, and hence, could serve as a resilience mechanism.

Tissue Chips and Microphysiological Systems
Tissue Chips (TCs) and Microphysiological Systems (MPSs) that incorporate human cells are novel platforms to model disease and screen drugs and provide an alternative to traditional animal studies. This review highlights the basic definitions of TCs and MPSs, examines four major organs/tissues, identifies critical parameters for organization and function (tissue organization, blood flow, and physical stresses), reviews current microfluidic approaches to recreate tissues, and discusses current shortcomings and future directions for the development and application of these technologies. The organs emphasized are those involved in the metabolism or excretion of drugs (hepatic and renal systems) and organs sensitive to drug toxicity (cardiovascular system). This article examines the microfluidic/microfabrication approaches for each organ individually and identifies specific examples of TCs. This review will provide an excellent starting point for understanding, designing, and constructing novel TCs for possible integration within MPS.

Proton radiobiology: DNA damage response
Clinical use of proton radiation has massively increased over the past years. The main reason for this is the beneficial depth-dose distribution of protons that allows to reduce toxicity to normal tissues surrounding the tumor. Despite the experience in the clinical use of protons, the radiobiology after proton irradiation compared to photon irradiation remains to be completely elucidated. Proton radiation may lead to differential damages and activation of biological processes.

Cerebral Ischemia
Cerebral ischemia represents the third cause of death and the first cause of disability in adults. This process results from decreasing cerebral blood flow levels as a result of the occlusion of a major cerebral artery. This restriction in blood supply generates low levels of oxygen and glucose, which leads to a decrease in the energy metabolism of the cell, producing inflammation, and finally, neurological deterioration. Currently, blood restoration of flow is the only effective approach as a therapy in terms of ischemic stroke. However, a significant number of patients still have a poor prognosis, probably owing to the increase in the generation of reactive oxygen species (ROS) during the reperfusion of damaged tissue. Oxidative stress and inflammation can be avoided by modulating mitochondrial function and have been identified as potential targets for the treatment of cerebral ischemia. In recent years, the beneficial actions of flavonoids and polyphenols against cerebrovascular diseases have been extensively investigated. The use of resveratrol (RSV) has been shown to markedly decrease brain damage caused by ischemia in numerous studies. According to in vitro and in vivo experiments, there is growing evidence that RSV is involved in several pathways, including cAMP/AMPK/SIRT1 regulation, JAK/ERK/STAT signaling pathway modulation, TLR4 signal transduction regulation, gut/brain axis modulation, GLUT3 up-regulation inhibition, neuronal autophagy activation, and de novo SUR1 expression inhibition.

Mirizzi Syndrome
Mirizzi syndrome is a rare condition caused by the compression of the common hepatic duct due to stones located in the cystic duct or the neck of the gallbladder, which causes obstruction of the extrahepatic biliary tract, what is most commonly presented as jaundice and upper abdominal pain. Mirizzi syndrome occurs approximately in 0.05-4% of patients undergiong cholecystectomy. Prolonged inflammation caused by the stones impacted in the cystic duct or the neck of the gallbladder may lead to advanced stages of Mirizzi syndrome and the formation of a cholecystocholedochal fistula or even a cholecystoenteric fistula. Diagnosis is made upon the symptoms, laboratory results and imaging techniques such as ultrasonography, computed tomography, magnetic resonance imaging or endoscopic retrograde choleangiopancreatography (ERCP), which is considered as the golden standard. However, the preoperative diagnosis is difficult and a large part of all cases is diagnosed intraoperatively. Management of Mirizzi syndrome is mostly surgical, but early stages of the syndrome can be treated with the use of ERCP.
