Triazolo-Thiadiazole Derivatives
The fusion of 1,2,4-triazole and 1,3,4-thiadiazole rings results in a class of heterocycles compounds with an extensive range of pharmacological properties. A series of 1,2,4-triazolo[3,4-b]-1,2,4-thiadiazoles was synthesized and tested for its enzyme inhibition potential and anticancer activity. The results show that 1,2,4-triazolo[3,4-b]-1,2,4-thiadiazoles display potent anticancer properties in vitro against a panel of cancer cells and in vivo efficacy in HT-29 human colon tumor xenograft in CB17 severe combined immunodeficient (SCID) mice.

Tremor in Parkinson’s Disease with Mechanical Devices
Parkinsonian tremors are sometimes confused with essential tremors or other conditions. Researchers conducted several studies on tremor evaluation using wearable sensors and devices, which may support an accurate diagnosis. Mechanical devices are also commonly used to treat tremors and have been actively researched and developed. Mechanical devices for tremor suppression include deep brain stimulation (DBS), electrical muscle stimulation, and orthosis. Adaptive DBS and optimization of stimulation parameters have been studied to improve treatment efficacy further. Due to developments using state-of-the-art techniques, effectiveness in diagnosing, evaluating, and suppressing tremors using these devices is satisfactorily high in many studies.

Tregs in Psoriasis
Psoriasis is a chronic inflammatory skin disease, which affects 2–4% of the population worldwide. Psoriasis has a strong genetic component, but environmental factors are also important. T-cell derived factors such as Psoriasis is characteristic, with thickening and scaling of the epidermisdue to hyper-proliferation of keratinocytes (acanthosis). CD4 and CD8 T-cell infiltrate characterizes the skin in psoriasis, a T-cell-driven disease, but other cell types are also present in the lesions, including neutrophils, macrophages, NK cells, dendritic cells. It has been long known that the pathogenesis of psoriasis is driven by T-cell derived factors, produced by subsets including T-helper (Th) 1 cells, Th17, Th22 and regulatory T cells (Tregs).

In December 2019, a patient was found to have pneumonia caused by an unknown betacoronavirus. With the unbiased sample sequencing of the patient, a novel coronavirus was identified and named Severe Acute Respiratory Syndrome Coronavirus 2, SARS-CoV-2, by the International Committee on Taxonomy of Viruses (ICTV).

Acute intermittent porphyria (AIP) is one of these rare diseases. AIP is characterized by haploinsufficiency in the third enzyme of the heme biosynthesis pathway. Identification of the liver as the target organ and a detailed molecular characterization have enabled the development and approval of several therapies to manage this disease.

Treatment Strategies of Epilepsy in Dravet Syndrome
Dravet Syndrome (DS) is a developmental epileptic encephalopathy characterized by drug-resistant seizures and other clinical features, including intellectual disability and behavioral, sleep, and gait problems. The pathogenesis is strongly connected to voltage-gated sodium channel dysfunction. The consensus of seizure management in DS consists of a combination of conventional and recently approved drugs such as stiripentol, cannabidiol, and fenfluramine. Despite promising results in randomized clinical trials and extension studies, the prognosis of the developmental outcomes of patients with DS remains unfavorable.

Treatment Strategies in Chronic Myeloid Leukaemia
Chronic myeloid leukaemia (CML) is a type of blood cancer that is currently well-managed with drugs that target cancer-causing proteins. However, a significant proportion of CML patients do not respond to those drug treatments or relapse when they stop those drugs because the cancer cells in those patients stop relying on that protein and instead develop a new way to survive. Therefore, new treatment strategies may be necessary for those patients.
