TRPV4 Ion Channel
The transient receptor potential vanilloid 4 channel (TRPV4) belongs to the mammalian TRP superfamily of cation channels. TRPV4 is ubiquitously expressed, activated by a disparate array of stimuli, interacts with a multitude of proteins, and is modulated by a range of post-translational modifications, the majority of which we are only just beginning to understand. Not surprisingly, a great number of physiological roles have emerged for TRPV4, as have various disease states that are attributable to the absence, or abnormal functioning, of this ion channel. This review will highlight structural features of TRPV4, endogenous and exogenous activators of the channel, and discuss the reported roles of TRPV4 in health and disease.

Mutations in TSC Genes and Synaptic Transmission
Tuberous sclerosis complex (TSC) is an autosomal dominant genetic disease featuring localized uncontrolled growth of tissues, known as hamartomas, in many organs, including the central nervous system (CNS). A large proportion of the cases are sporadic mutations (ca. two thirds). The disease results from a loss-of-function mutation in either the TSC1 or TSC2 gene, which encode for hamartin and tuberin proteins, respectively; two proteins that together form the Tsc1–Tsc2 complex. This complex, which is the converging center of several signaling pathways, has GTPase activity and can inhibit the activity of Ras homolog enriched in the brain (Rheb). The complex itself is modulated by phosphorylation via, for example, Akt or AMPK (Figure 1).

Peptide-Assisted Nucleic Acid Delivery Systems
Advances in peptide development have made peptide-assisted gene delivery more efficient in vitro and, in some instances, in small animal models. For example, cell and tissue selectivity could be greatly enhanced in the newest generation of CPPs. Other advances which allow for improved performance with regard to targeting and delivery of nucleic acids include adapting peptide sequences to facilitate escape or release from intracellular vesicles or respond to environmental stimuli for a controlled release of cargo, and the development of composite, multivalent peptide-based, or peptide-coupled structures.

SGLT2-Inhibitors on Epicardial Adipose Tissue
Sodium–glucose cotransporter-2 inhibitors (SGLT2-i) reduce adipose tissue and cardiovascular events in patients with type 2 diabetes (T2D). Accumulation of epicardial adipose tissue (EAT) is associated with increased cardio-metabolic risks and obstructive coronary disease events in patients with T2D. Studies suggest that the amount of EAT is significantly reduced in T2D patients with SGLT2-i treatment.

The Astrocytes in Brain Ischemia-Reperfusion Injury
Astrocytes account for 50% of the human brain volume and are normally classified into two mayor types according to morphological and spatial criteria: fibrous astrocytes in the white matter and protoplasmic astrocytes predominant in the grey matter. Astrocytes are the main glia of the central nervous system and play an important role both in brain physiology and in the response to damage. This article summarizes the most important evidence related to astrocytes and their response to cerebral ischemia.

Autophagy Modulation in Cholangiocarcinoma
Autophagy is a multistep catabolic process through which misfolded, aggregated or mutated proteins and damaged organelles are internalized in membrane vesicles called autophagosomes and ultimately fused to lysosomes for degradation of sequestered components. The multistep nature of the process offers multiple regulation points prone to be deregulated and cause different human diseases but also offers multiple targetable points for designing therapeutic strategies. Cancer cells have evolved to use autophagy as an adaptive mechanism to survive under extremely stressful conditions within the tumor microenvironment, but also to increase invasiveness and resistance to anticancer drugs such as chemotherapy.
